Virtue ethics is regarded as the key in search of moral excellence among corporations. Yet, there are limited works to empirically investigate what virtuous character morally good corporations is expected to exhibit in the course of business from the perspective of customers. To fill this gap, we argue that customers are to evaluate firm’s virtuous character using Confucian cardinal virtues (ren, yi, and li) and perceived virtuousness determines customer loyalty. We test this argument using a sample of 276 Hong Kong (...) Chinese. The result suggests that a corporation not only has to offer a good value and high quality to customers but also needs to develop and acquire virtuous character in retaining customer loyalty. Firm has to embrace ren, yi, and li as the primary business goal. In fact, they are not the means but the end in themselves. The continuous pursuit of these cardinal virtues at the firm level not only establishes proper corporate values but also enables a firm to act morally in the course of business. TanakaAs the title of my paper indicates, two features of my identity have become more vivid as the result of my participation in the International Buddhist-Christian Theological Encounter (IBCTE) sessions. The first of the two stemmed from my rude awakening that not everyone involved with our (...) sessions accepted the assumption that the other was a similarly valid soteriological path.In one of our sessions a few years back, the discussion moved to a topic of the nature of "ultimate path," which, as it turned out, began to raise some extremely sensitive issues. An unusually tense atmosphere hung over the discussion table, a rare turn of events for our usually jovial dialogue group composed mostly of liberal-minded members. One of the Christian members gave voice to her fundamental view that Jesus was the sole path through which one can be saved. It was clearly what anyone would regard as an exclusivist position. In asking for further clarification, she reiterated her view that her Christian faith necessitated that she took that position, and that I as a Buddhist was, therefore, not included in that soteriological scheme. I responded to her that I understood that I would not be in her scheme since I was not Christian, but would I nevertheless be saved? After some hesitation, in so many words, she replied that I would not be! Had she been a member of a more conservative group, it would have been easier to accept that kind of a response, but coming from an academic with liberal leanings, her comments caught me off guard. But on the other hand, I appreciated her honesty in being true to her understanding of the tradition, for it seemed to me that it was not an easy thing for her to say.This short encounter has been etched firmly in my mind and, ironically, remains for me the most memorable moment among all of the IBCTE sessions attended. I did not pursue the matter further at that time (aside from the fact that time had run [End Page 41] out for that particular session), since I was aware that such an exclusive position (as distinguished from those of inclusivism and pluralism) was common among Christians. I just happened to have encountered it in a rather unexpected circumstance and was not prepared with a comeback.This encounter forced me to inquire about my own position concerning the salvation of people other than Buddhists. Would I regard her as not being saved? After some probing and reflection on my part, I can say with little hesitation that I would never be able to say, "no, you are not." I have no basis on which to refute what her time-tested tradition promises. I am not in a position to negate the claims of her tradition or to question the sincerity of her commitment. From my Buddhist perspective there is no universal criterion on which to make judgments about others. TanakaIn his celebrated book Bowling Alone (2000), Robert Putnam noted the increased level in the phenomenon of "privatized religion" within the previous thirty-five years. Many of the Baby Boomer generation left churches in the late 1960s and the 1970s. Some sought out new religious movements and religious therapies, but most simply "dropped out" (...) of organized religion altogether. He cites Wade Clark Roof and William McKinney, specialists in American religion, who describe the outcome of this trend in American religious behavior: "[The consequence was a] tendency toward highly individualized religious psychology without the benefits of strong supportive attachments to believing communities. A major impetus in this direction in the post-1960s was the thrust toward greater personal fulfillment and quest for the ideal self.... In this climate of expressive individualism, religion tends to become "privatized," or anchored in the personal realms."1 Putnam observes that although privatized religion may be "morally compelling and psychologically fulfilling," it involves less social capital than environments in which individuals are connected to other individuals in shared faith commitment.Interestingly, American Buddhism may be one of the beneficiaries of this trend, for the recent growth of Buddhism in the United States is partly attributable to the climate of "privatized" religion. Nowhere is this symbolized more than by the so-called Nightstand Buddhists.2The Nightstand Buddhists derive their name from their practice of placing a Buddhist meditation book on the nightstand after reading it before they go to sleep. They get up the next morning and practice to the best of their ability the meditation they had read about the night before. Further, they may frequently attend lectures at the local university and visit a Buddhist center's web-page or participate in an online Buddhist discussion group. And if we were to visit them, we might find their homes decorated with Buddhist artifacts.But these Nightstand Buddhists do not show up in any statistics on the [End Page 115] American Buddhist population because they would not identify themselves as Buddhists. According to Thomas Tweed, who coined the term Nightstand Buddhists, "Sympathizers (= Nightstand Buddhists) are those who have some sympathy for Buddhism but do not embrace it exclusively or fully. When asked, they would not identify themselves as Buddhists. They would say they are Methodists, or Jewish, or unaffiliated."3 However, they have been an important part of the story of Buddhism in America from its beginning and continue to be so today.Among the three million Buddhists currently estimated to be in the United States, I believe there are Buddhists who exhibit many of the same characteristics as the Nightstand Buddhists. And this number is significant, particularly among those converts to Buddhism whose main practice is meditation. This is an interesting question in itself. I personally don't know of anyone who fits that description or who has actually witnessed someone like her in prayer. Nevertheless, it is interesting how a Buddhist raised in a predominately Christian society comes to form his image of Christian prayer.Working further with this image, I see this little girl—let's call her Megan—doing two things in her evening prayer. One is that she is giving thanks to God for the benefits she has received. She thanks him for her food, clothing, friends, and family. Then, she also makes a request. In this petitionary element of her prayer, she is asking God to make her sick father get well soon. As she does so, her expression turns more serious, and she clasps her hands more firmly and brings them closer to her chest. She then looks up toward the ceiling, expressing her heartfelt wish for her father's early recovery. What is noteworthy is that Megan's prayers are spoken aloud as in a human conversation.I realize that this image does not do justice to a topic as enormous as "Christian prayer," but I believe that it provides a glimpse into how one Buddhist imagines the topic. I have a number of questions about Megan's prayer, particularly as I try to understand what may be going on inside of her. I wonder first and foremost, what God looks like to her. Is God a male or a female, or neither? I would think that even for her (in this gender-sensitive age), God is still a male. If so, does she see him as an elderly man with a long white beard, holding a cane and sitting in a throne? Next, where does God reside? If he is in heaven, what is heaven like to her? I wonder if heaven is located somewhere amidst the lofty, puffy clouds. In regards to her father's illness, how does she think God can make her father get well? What is the mechanism by which a personified God can heal? Perhaps I am being too analytical and overly rigorous with my questions about a spiritual act, and that of an eight-year-old at that.Perhaps I should, instead, turn the questions toward me, asking how I would [End Page 87] answer the same questions about a Buddhist girl "praying" before the Buddha or a bodhisattva for the same kind of wish? The image that immediately crops up in my mind is of a girl of about twelve and her mother whom I saw at a pagoda in Rangoon, Burma, a number of years ago. The girl is wearing a large, red fresh flower in her hair and has on traditional apparel. She sits with her mother on a marble floor of a spacious area in front of Buddha images that are several meters in height. She sits with her legs folded but thrown out to her side, a style common in her country as well as in Thailand. Her eyes appear closed, and her head is slightly bowed down toward the Buddha images. Heidelberg: Universitätsverlag, 2006. 156 pp.In Asura’s Harp, Hirota focuses on the Pure Land Buddhist thought of Shinran (1173–1263), the founder of Jōdo Shinshō school and one of the major figures of Japanese Buddhism. I believe Hirota’s main argument of the book is succinctly expressed on its back cover (...) blurb: “Unlike monastic traditions that seek the eradication of delusional attachments through transcendence of ordinary thought and speech, Shinran teaches that human existence is inevitably characterized by linguisticality and that the genuinely practicable path to realization lies in engagement with language.” For Hirota, language plays a central role in the religious transformative experience called Shinjin, considered a genuine awareness of the self and the world. This way of understanding religious language goes against the dominant view, especially that of Zen with its proverbial “beyond words” approach 1 and contemporary Buddhist discussions on language as being limited and even false.In the prologue of the book, Hirota points out the growing convergence in the Buddhist outlook and the current interest in Western thought. Both traditions see that “persons are not autonomous agents making judgments and initiating actions independently of others, their past, and their location in society and history” (p. 2). Instead, humans are now seen to be situated in particular temporal and spatial circumstances, with its attendant limitations, prejudices, and finitude. In terms of language, this realization has led to a shift in Western thought from metaphysical to hermeneutical concerns, thus resonating with Buddhism’s view of ordinary language [End Page 182] as “inevitably warped by attachments and informed by falsely reifying conceptualization” (p. 2).At the same time both traditions recognize the adaptive use of language, as seen especially in the Buddhist conception of “skillful means,” to provide guidance to self-realization and awareness. This, Hirota believes, offers an opportunity for a dialogue between contemporary Western thought and Buddhism.With this as the backdrop, Hirota sets out to clarify the nature and significance of language in Shin Pure Land thought with an intent to contribute to this broader discussion of religious language across religious and cultural boundaries, a formidable task for any scholar. However, there is no one, in my opinion, more qualified to carry out this task than Hirota, for he served as the chief translator of the twenty-year project that culminated in the Collected Works of Shinran (CWS)2 and has been publishing insightful and provocative writings in the study of language from Shin Buddhist perspective in both English and Japanese.3 There are very few, if any, scholars in Japan dealing with this subject with the ability to talk about it in a cross-religious and disciplinary context. In more recent years, he has taken his findings to gatherings outside Japan, particularly to Europe. In fact, this book is, in large part, an outcome of the talks he delivered several years ago in the Fürst Franz-Josef and Fürstin Gina Memorial Philosophy Lecture Series at the Internationale Akademie für Philosophie in Liechtenstein.Before I focus in on some of the central points in this book, it would be helpful to get a bird’s eye view of this book, Asura’s Harp by listing the chapter titles. Over the past two decades, Victor and Cullen's (Adm Sci Q 33:101-125, 1988) typology of ethical climates has been employed by many academics in research on issues of ethical climates. However, little is known about how managerial practices such as communication and empowerment influence ethical climates, especially from a functional perspective. The current study used a survey of employees from Taiwan's top 100 patent-owning companies to examine how communication and empowerment affect organizational ethical climates. The results confirm the relationship between (...) these two managerial practices and organizational ethical climates. Ethical sensitivity triggers the entire ethical decision-making process (i.e., recognition of ethical content in work situations). In this article, five factors are examined that affect tax practitioners' professional ethical sensitivity. The five factors that were examined include role conflict, role ambiguity, job satisfaction, professional commitment, and ethical orientation. Ethical content in work situations is examined in relation to professional ethics as enumerated by the American Institute of Certified Public Accountant's (AICPA) Statements on Responsibilities in Tax Practice (SRTP). Utilizing Hunt and (...) Vitell's (1986, 1993) General Theory of Ethics, a model of ethical sensitivity was constructed and empirically tested. Role conflict negatively and job satisfaction positively influenced tax practitioners' ethical sensitivity. Also, the covariates of the tax practitioner's professional risk level and type of employer were found to be significant. The significant factors are job specific. The tax firm may have the best opportunity to positively change a tax practitioner's ethical recognition abilities. Professional accounting organizations may need to evaluate if resources should be used to formulate, maintain, and publicize codes of conduct because of the lack of significance of professional commitment. Event-related desynchronization, as a proxy for mirror neuron activity, has been used as a neurophysiological marker for motor execution after mirror visual feedback. Using EEG, this study investigated ERD upon the immediate effects of single-session MVF in unimanual arm movements compared with the ERD effects occurring without a mirror, in two groups: stroke patients with left hemiplegia and their healthy counterparts. During EEG recordings, each group performed one session of mirror therapy training in three task conditions: with a mirror, with (...) no mirror, and with a covered mirror. An asymmetry index was calculated from the subtraction of the event-related spectrum perturbations between the C3 and C4 electrodes located over the sensorimotor cortices contralateral and ipsilateral to the moved arm. Results of the effect of task versus group in contralateral and ipsilateral motor areas showed that there was a significant effect of task condition at the contralateral motor area in the high beta band at C3. High beta ERD showed that the suppression was greater over the contralateral hemisphere than it was over the ipsilateral hemisphere in both study groups. The magnitude of low beta ERD in patients with stroke was more suppressed in contralesional C3 under the no mirror compared to that of the covered mirror and similarly more suppressed in ipsilesional C4 ERD under the no mirror compared to that of the mirror condition. The correlation analysis revealed that the magnitude of ERSP power correlated significantly with arm severity in the low and high beta bands in patients with stroke, and a higher asymmetry index in the low beta band was associated with higher arm functioning under the no-mirror condition. There was a shift in sensorimotor ERD toward the contralateral hemisphere as induced by MVF accompanying unimanual movement in both stroke patients and healthy controls. The use of ERD in the low beta band as a neurophysiological marker to indicate the relationships between the amount of MVF-induced ERD attenuation and motor severity, and the outcome indicator for improving stroke patients’ neuroplasticity in clinical trials using MVF are warranted to be explored in the future. As studies continue to accumulate on leader humility, it has become clear that humility in a leader is largely beneficial to his or her followers. While the majority of the empirical research on this topic has demonstrated the positive effects of leader humility, this study challenges that consensus by arguing that a leader’s humble behavior can have contradictory outcomes in followers’ voice behavior. Drawing on attachment theory, we develop a model which takes into account the ways in which leader humility (...) influences the seemingly contradictory voice behavior of followers, i.e., inducing challenging voice, and defensive voice depending on the followers’ sense of security as reflected by feeling trusted and self-efficacy for voice. The results of this empirical study confirm that leader humility influences followers’ voice in a contradictory way through their sense of security. (shrink)

The apolipoprotein E gene plays an important role in the pathogenesis of Alzheimer's disease , and amyloid plaque comprised mostly of the amyloid-beta peptide ) is one of the major hallmarks of AD. However, the relationship between these two important molecules is poorly understood. We examined how A treatment affects APOE expression in cultured cells and tested the role of the transcription factor NF-B in APOE gene regulation. To delineate NF-B's role, we have characterized a 1098 nucleotide segment containing the (...) 5'-flanking region of the human APOE gene . Sequence analysis of this region suggests the presence of two potential NF-B elements. To demonstrate promoter activity, the region was cloned upstream of a promoterless luciferase gene. This segment was able to drive expression of luciferase in transient transfections of human fetal glial cells. Promoter activity was stimulated twofold by A treatment. Pretreatment with double-stranded DNA decoy oligonucleotides against NF-B reduced A stimulation. Deletion and mutagenetic analyses demonstrated that the distal NF-B element was functional and showed a strong DNA-protein complex band in gel shift analysis, similar to that from control NF-B consensus element. An anti-inflammatory and anti-NF-B drug, sodium salicylate, significantly blocked A-induced APOE promoter function. Our data provide evidence that upregulation of APOE by A in astroglial cells is mediated by an NF-B-element present in the 5'-flanking region of the APOE gene. (shrink)

Replication protein A (RPA) is the main eukaryotic single‐stranded DNA (ssDNA) binding protein, having essential roles in all DNA metabolic reactions involving ssDNA. RPA binds ssDNA with high affinity, thereby preventing the formation of secondary structures and protecting ssDNA from the action of nucleases, and directly interacts with other DNA processing proteins. Here, we discuss recent results supporting the idea that one function of RPA is to prevent annealing between short repeats that can lead to chromosome rearrangements by microhomology‐mediated end (...) joining or the formation of hairpin structures that are substrates for structure‐selective nucleases. We suggest that replication fork catastrophe caused by depletion of RPA could result from cleavage of secondary structures by nucleases, and that failure to cleave hairpin structures formed at DNA ends could lead to gene amplification. These studies highlight the important role RPA plays in maintaining genome integrity. (shrink)

The purpose of this study was to develop a Physician's Spiritual Well-Being Scale (PSpWBS). The significance of a physician's spiritual well-being was explored through in-depth interviews with and qualitative data collection from focus groups. Based on the results of qualitative analysis and related literature, the PSpWBS consisting of 25 questions was established. Reliability and validity tests were performed on 177 subjects. Four domains of the PSpWBS were devised: physician's characteristics; medical practice challenges; response to changes; and overall well-being. The explainable (...) total variance was 65.65%. Cronbach α was 0.864 when the internal consistency of the whole scale was calculated. Factor analysis showed that the internal consistency Cronbach α value for each factor was between 0.625 and 0.794 and the split-half reliability was 0.865. The scale has satisfactory reliability and validity and could serve as the basis for assessment of the spiritual well-being of a physician. (shrink)

The present study investigated whether another individual’s gaze direction influences an observer’s affective responses. In Experiment 1, subjective self-ratings and an affective priming paradigm were employed to examine how participants explicitly and implicitly, respectively, evaluated the affective valence of direct gaze, averted gaze, and closed eyes. The explicit self-ratings showed that participants evaluated closed eyes more positively than direct gaze. However, the implicit priming task showed an inverse pattern of results indicating that direct gaze was automatically evaluated more positively than (...) closed eyes were. Experiment 2 confirmed that the opposite patterns of results between the two tasks were not due to differences in presentation times of the gaze stimuli. The results provide evidence for automatic affective reactions to eye gaze and indicate a dissociation between explicit and implicit affective evaluations of eyes and gaze direction. (shrink)

It is often thought that epistemic relations between experience and belief make it possible for our beliefs to be about or "directed towards" the empirical world. I focus on an influential attempt by John McDowell to defend a view along these lines. According to McDowell, unless experiences are the sorts of things that can be our reasons for holding beliefs, our beliefs would not be "answerable" to the facts they purportedly represent, and so would lack all empirical content. I argue (...) that there is no intelligible conception of what it is for beliefs to be answerable to the facts that supports McDowell's claim that our empirical beliefs must be justified by experience. Objectives: To quantify the use of do-not-resuscitate orders in a tertiary-care children’s hospital and to characterise the circumstances in which such orders are written.Design: Retrospective study conducted in a 500-bed children’s hospital in Taiwan.Patients: The course of 101 patients who died between January 2002 and December 2005 was reviewed. The following data were collected: age at death, gender, disease and its status, place of death and survival. There were 59 males and 42 females with a median age of 103 months (...) . 50 children had leukaemias, and 51 had malignancies other than leukaemia. The t test and the χ2 test were applied as appropriate.Results: The study found that 44% of patient deaths occurred in the paediatric oncology ward; 29% of patient deaths occurred in the intensive care unit; and 28% of patients died in their home or at another hospital. Other findings included the following: 46 of 101 patients died after attempted cardiopulmonary resuscitation and 55 died with a DNR order in effect. The mean age at death was 9.8 years in both groups with or without DNR orders.Conclusions: From the study of patient deaths in this tertiary-care children’s hospital, it was concluded that an explicit DNR order is now the rule rather than the exception, with more DNR orders being written for patients who have been ill longer, who have solid tumours, who are not in remission and who are in the ward. Monetary intelligence theory asserts that individuals apply their money attitude to frame critical concerns in the context and strategically select certain options to achieve financial goals and ultimate happiness. This study explores the dark side of monetary Intelligence and behavioral economics—dishonesty. Dishonesty, a risky prospect, involves cost–benefit analysis of self-interest. We frame good or bad barrels in the environmental context as a proxy of high or low probability of getting caught for dishonesty, respectively. We theorize: The magnitude and intensity of (...) the relationship between love of money and dishonest prospect may reveal how individuals frame dishonesty in the context of two levels of subjective norm—perceived corporate ethical values at the micro-level and Corruption Perceptions Index at the macro-level, collected from multiple sources. Based on 6382 managers in 31 geopolitical entities across six continents, our cross-level three-way interaction effect illustrates: As expected, managers in good barrels, mixed barrels, and bad barrels display low, medium, and high magnitude of dishonesty, respectively. With high CEV, the intensity is the same across cultures. With low CEV, the intensity of dishonesty is the highest in high CPI entities —the Enron Effect, but the lowest in low CPI entities. CPI has a strong impact on the magnitude of dishonesty, whereas CEV has a strong impact on the intensity of dishonesty. We demonstrate dishonesty in light of monetary values and two frames of social norm, revealing critical implications to the field of behavioral economics and business ethics. (shrink)

RAID-II is a high-bandwidth, network-attached storage server designed and implemented at the University of California at Berkeley. In this paper, we measure the performance of RAID-II and evaluate various architectural decisions made during the design process. We first measure the end-to-end performance of the system to be approximately 20 MB/s for both disk array reads and writes. We then perform a bottleneck analysis by examining the performance of each individual subsystem and conclude that the disk subsystem limits performance. By adding (...) a custom interconnect board with a high-speed memory and bus system and parity engine, we are able to achieve a performance speedup of 8 to 15 over a comparative system using only off-the-shelf hardware. Corruption involves greed, money, and risky decision-making. We explore the love of money, pay satisfaction, probability of risk, and dishonesty across cultures. Avaricious monetary aspiration breeds unethicality. Prospect theory frames decisions in the gains-losses domain and high-low probability. Pay dissatisfaction (in the losses domain) incites dishonesty in the name of justice at the individual level. The Corruption Perceptions Index, CPI, signals a high-low probability of getting caught for dishonesty at the country level. We theorize that decision-makers adopt avaricious love-of-money aspiration (...) as a lens and frame dishonesty in the gains-losses domain (pay satisfaction-dissatisfaction, Level 1) and high-low probability (CPI, Level 2) to maximize expected utility and ultimate serenity. We challenge the myth: Pay satisfaction mitigates dishonesty across nations consistently. Based on 6500 managers in 32 countries, our cross-level three-dimensional visualization offers the following discoveries. Under high aspiration conditions, pay dissatisfaction excites the highest- (third-highest) avaricious justice-seeking dishonesty in high (medium) CPI nations, supporting the certainty effect. However, pay satisfaction provokes the second-highest avaricious opportunity-seizing dishonesty in low CPI entities, sustaining the possibility effect—maximizing expected utility. Under low aspiration conditions, high pay satisfaction consistently leads to low dishonesty, demonstrating risk aversion—achieving ultimate serenity. We expand prospect theory from a micro and individual-level theory to a cross-level theory of monetary wisdom across 32 nations. We enhance the S-shaped Curve to three 3-D corruption surfaces across three levels of the global economic pyramid, providing novel insights into behavioral economics, business ethics, the environment, and responsibility. The embodied simulation of smiles involves motor activity that often changes the perceivers' own emotional experience (e.g., smiling can make us feel happy). Although Niedenthal et al. mention this possibility, the psychological processes by which embodiment changes emotions and their consequences for processing other emotions are not discussed in the target article's review. We argue that understanding the processes initiated by embodiment is important for a complete understanding of the effects of embodiment on emotion perception.

Access to legal information is fundamental to access to justice. Yet accessibility refers not only to making legal documents available to the public, but also rendering legal information comprehensible to them. A vexing problem in bringing legal information to the public is how to turn formal legal documents such as legislation and judgments, which are often highly technical, to easily navigable and comprehensible knowledge to those without legal education. In this study, we formulate a three-step approach for bringing legal knowledge (...) to laypersons, tackling the issues of navigability and comprehensibility. First, we translate selected sections of the law into snippets (called CLIC-pages), each being a small piece of article that focuses on explaining certain technical legal concept in layperson’s terms. Second, we construct a _Legal Question Bank_, which is a collection of legal questions whose answers can be found in the CLIC-pages. Third, we design an interactive _CLIC Recommender_. Given a user’s verbal description of a legal situation that requires a legal solution, CRec interprets the user’s input and shortlists questions from the question bank that are most likely relevant to the given legal situation and recommends their corresponding CLIC pages where relevant legal knowledge can be found. In this paper we focus on the technical aspects of creating an LQB. We show how large-scale pre-trained language models, such as GPT-3, can be used to generate legal questions. We compare machine-generated questions against human-composed questions and find that MGQs are more scalable, cost-effective, and more diversified, while HCQs are more precise. We also show a prototype of CRec and illustrate through an example how our 3-step approach effectively brings relevant legal knowledge to the public. (shrink)

COVID-19 often manifests with different outcomes in different patients, highlighting the complexity of the host-pathogen interactions involved in manifestations of the disease at the molecular and cellular levels. In this paper, we propose a set of postulates and a framework for systematically understanding complex molecular host-pathogen interaction networks. Specifically, we first propose four host-pathogen interaction (HPI) postulates as the basis for understanding molecular and cellular host-pathogen interactions and their relations to disease outcomes. These four postulates cover the evolutionary dispositions involved (...) in HPIs, the dynamic nature of HPI outcomes, roles that HPI components may occupy leading to such outcomes, and HPI checkpoints that are critical for specific disease outcomes. Based on these postulates, an HPI Postulate and Ontology (HPIPO) framework is proposed to apply interoperable ontologies to systematically model and represent various granular details and knowledge within the scope of the HPI postulates, in a way that will support AI-ready data standardization, sharing, integration, and analysis. As a demonstration, the HPI postulates and the HPIPO framework were applied to study COVID-19 with the Coronavirus Infectious Disease Ontology (CIDO), leading to a novel approach to rational design of drug/vaccine cocktails aimed at interrupting processes occurring at critical host-coronavirus interaction checkpoints. Furthermore, the host-coronavirus protein-protein interactions (PPIs) relevant to COVID-19 were predicted and evaluated based on prior knowledge of curated PPIs and domain-domain interactions, and how such studies can be further explored with the HPI postulates and the HPIPO framework is discussed. The sporadic nature of Alzheimer's disease argues for an environmental link that may drive AD pathogenesis; however, the triggering factors and the period of their action are unknown. Recent studies in rodents have shown that exposure to lead during brain development predetermined the expression and regulation of the amyloid precursor protein and its amyloidogenic beta-amyloid product in old age. Here, we report that the expression of AD-related genes [APP, BACE1 ] as well as their transcriptional regulator were elevated in aged (...) monkeys exposed to Pb as infants. Furthermore, developmental exposure to Pb altered the levels, characteristics, and intracellular distribution of Abeta staining and amyloid plaques in the frontal association cortex. These latent effects were accompanied by a decrease in DNA methyltransferase activity and higher levels of oxidative damage to DNA, indicating that epigenetic imprinting in early life influenced the expression of AD-related genes and promoted DNA damage and pathogenesis. These data suggest that AD pathogenesis is influenced by early life exposures and argue for both an environmental trigger and a developmental origin of AD. (shrink)
