Multigene delivery and expression systems are emerging as key technologies for many applications in contemporary biology. We have developed new methods for multigene delivery and expression in eukaryotic hosts for a variety of applications, including production of protein complexes for structural biology and drug development, provision of multicomponent protein biologics, and cell‐based assays. We implemented tandem recombineering to facilitate rapid generation of multicomponent gene expression constructs for efficient transformation of mammalian cells, resulting in homogenous cell populations. Analysis of multiple parameters (...) in living cells may require co‐expression of fluorescently tagged sensors simultaneously in a single cell, at defined and ideally controlled ratios. Our method enables such applications by overcoming currently limiting challenges.Here, we review recent multigene delivery and expression strategies and their exploitation in mammalian cells. We discuss applications in drug discovery assays, interaction studies, and biologics production, which may benefit in the future from our novel approach. (shrink)

Proofs and Refutations is essential reading for all those interested in the methodology, the philosophy and the history of mathematics. Much of the book takes the form of a discussion between a teacher and his students. They propose various solutions to some mathematical problems and investigate the strengths and weaknesses of these solutions. Their discussion raises some philosophical problems and some problems about the nature of mathematical discovery or creativity. Imre Lakatos is concerned throughout to combat the classical picture (...) of mathematical development as a steady accumulation of established truths. He shows that mathematics grows instead through a richer, more dramatic process of the successive improvement of creative hypotheses by attempts to 'prove' them and by criticism of these attempts: the logic of proofs and refutations. Volume II presents his work on the philosophy of mathematics (much of it unpublished), together with some critical essays on contemporary philosophers of science and some famous polemical writings on political and educational issues. Imre Lakatos had (...) an influence out of all proportion to the length of his philosophical career. This collection exhibits and confirms the originality, range and the essential unity of his work. It demonstrates too the force and spirit he brought to every issue with which he engaged, from his most abstract mathematical work to his passionate 'Letter to the director of the LSE'. This paper proposes a reading of Kelsen’s basic norm based on Kant’s regulative ideas. I begin by exposing Kant’s conception of the principles of reason. Then I criticize an interpretation of the basic norm along the same lines made by Stanley Paulson. Thirdly I analyze two theses from Hermann Cohen that influenced Kelsen greatly and reinforce my stance on the basic norm. Lastly, I explain how the Kelsenian tenet that the basic norm is the transcendental grounding of the normativity of (...) law can be comprehended based on the role the ideas of reason play in the systematization of knowledge. Many have claimed that epistemic rationality sometimes requires us to have imprecise credal states (i.e. credal states representable only by sets of credence functions) rather than precise ones (i.e. credal states representable by single credence functions). Some writers have recently argued that this claim conflicts with accuracy-centered epistemology, i.e., the project of justifying epistemic norms by appealing solely to the overall accuracy of the doxastic states they recommend. But these arguments are far from decisive. In this essay, we prove some (...) new results, which show that there is little hope for reconciling the rationality of credal imprecision with accuracy-centered epistemology. (shrink)

All societies are made up of members who have a certain number of things in common, by virtue of which they understand, identify and communicate with each other, on the one hand, and establish differences with members of other societies on the other. Among the most important of these things is language.In this regard, let us recall two examples raised by Ferdinand de Saussure. The first: “Language is both a social product of the faculty of speech and an ensemble of (...) necessary conventions adopted by the social body to allow for the exercise of this faculty in each individual.” The second: “Speech, distinct from language, is on the contrary an individual act of intelligence and will … by which the speaking subject uses the code of language to express his personal thought.”. Isolated habitats can be compared and ranked by comparing their interior-to-edge ratio (I/E). We would like to show here that results based on ranking by I/E ratio sometimes contradict Diamond's rule, which ranks the most rounded habitat (i.e. most compact) as the best one. The reason for this contradiction is the frequently overlooked size dependence of the I/E. Being the interior-to-edge ratio size dependent, from a given set of habitats of different sizes, compact shaped (rounded) habitats might have worse I/E (...) ratios than elongated or irregular ones.
