We discuss here a series of testable hypotheses concerning the role of chromosome folding into topologically associating domains (TADs). Several lines of evidence suggest that segmental packaging of chromosomal neighborhoods may underlie features of chromatin that span large domains, such as heterochromatin blocks, association with the nuclear lamina and replication timing. By defining which DNA elements preferentially contact each other, the segmentation of chromosomes into TADs may also underlie many properties of long‐range transcriptional regulation. Several observations suggest that TADs can (...) indeed provide a structural basis to regulatory landscapes, by controlling enhancer sharing and allocation. We also discuss how TADs may shape the evolution of chromosomes, by causing maintenance of synteny over large chromosomal segments. Finally we suggest a series of experiments to challenge these ideas and provide concrete examples illustrating how they could be practically applied. (shrink)

The earliest ancient Greek text to narrate the resolution of a large-scale conflict by judicial means is Aeschylus's tragedy Eumenides, first performed in Athens in 458 BC. After explaining the historical context in which the play was performed—a context of acute civic discord and the imminent danger of an escalation of reciprocal revenge killings by the lower-class faction in Athens—this article offers a new reading of the play and asks if it can help us think about the challenges inherent in (...) conflict resolution today. The prosecutors are the Erinyes, the archaic supernatural agents of murder victims; their responsibility, in predemocratic Greece, before the invention of law courts, was to punish murderers. The defendant is Orestes, who has killed his mother but argues that, since she had killed his father, he was acting justly. The judges consist of eleven Athenian citizen jurors plus the presiding god, Athena, whose twelfth vote carries slightly more weight than any of the others. The article concludes that some aspects of the procedure are exemplary, especially Athena's insistence on respecting each side's right to be heard, her sensitivity toward the grievances felt by the defeated Erinyes, and the compensation they are offered. On the other hand, the tragedy clearly shows how difficult it is for a fair legal judgment to be made without a view to larger issues of national expedience, security, and inherent power structures, especially that of patriarchy. This initial volume of the Collected Works of Edith Stein offers, for the first time in English, the unabridged biography of Edith Stein (Teresa Benedicta of the Cross), depicting her life as a child and young adult. Her text ends abruptly because the Nazi SS arrested, then deported, her to the Auschwitz concentration camp in 1942. Edith Stein is one of the most significant German women of the 20th century. At the age of twenty-five she became the (...) first assistant to Edmund Husserl, the founder of Phenomenology. She was much in demand as a writer and lecturer after her conversion from atheism to Catholicism. Later, as a Discalced Carmelite nun, she maintained her intellectual pursuits until she, like so many others, became a victim of the Nazi persecution that raged across Eastern Europe. By making this landmark work available in English, the Institute of Carmelite Studies provides an eye-witness account of persons and activities on the scene at the time when psychology and philosophy became separate disciplines. In addition to photographs and a map, this volume is enhanced with a preface, the foreword and afterword, notes, and a list of places associated with Edith Stein’s life. Edith Russell had already written the lives of Carey Thomas and Wilfrid Scawen Blunt when she married Bertrand Russell in 1952. She preserved his files as no one before had, and took a great interest in his earlier years as she did in his current campaigns and family. When Clark’s Life appeared in 1975, she reacted strongly to it. She wrote three drafts of her comments, each draft more extensive, and including information only she would have, such as Russell’s (...) views on personal matters, even his habits of dress. Electoral control refers to attempts by an election's organizer to influence the outcome by adding/deleting/partitioning voters or candidates. The important paper of Bartholdi, Tovey, and Trick [1] that introduces control proposes computational complexity as a means of resisting control attempts: Look for election systems where the chair's task in seeking control is itself computationally infeasible.We introduce and study a method of combining two or more candidate-anonymous election schemes in such a way that the combined scheme possesses all the resistances to (...) control possessed by any of its constituents: It combines their strengths. From this and new resistance constructions, we prove for the first time that there exists a neutral, anonymous election scheme that is resistant to all twenty standard types of electoral control. Max Weber died suddenly on 14 June 1920 in Munich mid-way through a course of lectures on “Sociology of the State” and with two books in the press: the three volumes of his sociology of religion, and the first four chapters of Wirtschaft und Gesellschaft. His widow, Marianne Weber, at the time active in regional politics and the women's movement, immediately suspended all commitments other than work on her dead husband's intellectual legacy. In the following years she completed Wirtschaft und (...) Gesellschaft from manuscripts, edited four volumes of his collected writings, assembled Max Weber's extensive correspondence, and in 1926 completed a definitive biography. No other thinker so engaged the Russian cultural imagination of the early twentieth century as did Friedrich Nietzche. The Revolution of Moral Consciousness shows how Nietzschean thought influenced the brilliant resurgence of literary life that started in the 1890s and continued for four decades. Through an analysis of the Russian encounter with Nietzsche, Edith Clowes defines the shift in ethical and aesthetic vision that motivated Russia's unprecedented artistic renascence and at the same time led its followers to the brink (...) of cultural despair. Clowes shows how in the last years of the nineteenth century a diverse array of writers and critics discovered Nietzsche's thought, embracing or repudiating it with equal vigor. The literary storm brewing around Nietzsche and the concurrent relaxation of censorship combined to attract a public eager to follow the new intellectual fashion. Young writers, such as Andreev and Kuprin, welcomed the idea of the "superman" as a promising path to personal fulfillment. The tragic fates of their protagonists and the alluring gospel of the vulgar Zarathustra-like characters of such bestselling authors as Boborykin, Artsybashev, and Verbitskaia found enthusiastic, if indiscriminating, audiences ready to be "taught" how to "find themselves." By considering this Nietzschean cult, Clowes draws fresh insight into the nature of the budding popular-culture industry in Russia and the fast-growing reading public. From this ferment emerged the greatest Russian literary voices of the early twentieth century. The revolutionary romantics, Gorky and Lunacharsky, sought in Nietzsche's writing a new vision of total social and cultural change. Merezhkovsky led a generation of mystic symbolists in the search for a literary myth of resurrection. Ivanov, Blok, and Belyi appropriated the image of the "crucified Dionysus" as the central symbol of spiritual transfiguration. Their encounters with Nietzschean thought disclose an even more profound creative struggle with their own cultural past and its established formulations of nation and individual, culture and history. Clowes uses the term future anxiety to speak of a creative mentality that strove to assert itself by diminishing the impact of powerful literary precursors, such as Tolstoi, Dostoevsky, and Solovyov, and opening to the imagination the vision of a future full of vast creative possibility. We investigate the effect on the complexity of adding the universal modality and the reflexive transitive closure modality to modal logics. From the examples in the literature, one might conjecture that adding the reflexive transitive closure modality is at least as hard as adding the universal modality, and that adding either of these modalities to a multi-modal logic where the modalities do not interact can only increase the complexity to EXPTIME-complete. We show that the first conjecture holds under reasonable assumptions (...) and that, except for a number of special cases which we fully characterize, the hardness part of the second conjecture is true. However, the upper bound part of the second conjecture fails miserably: we show that there exists a uni-modal, decidable, finitely axiomatizable, and canonical logic for which adding the universal modality causes undecidability and for which adding the reflexive transitive closure modality causes high undecidability. This chapter uses feminist standpoint theory to investigate Diotima’s epistemic advantage in Plato’s Symposium. Scholars have wondered why Diotima – a woman speaking about the role of erōs in gestation, childbirth, and childrearing – voices the view that Plato privileges most among all the symposiasts (Halperin 1990, Evans 2006, Hobbs 2007). Feminist standpoint theory is useful in developing a novel answer to this question; it supposes that oppressed groups, because they occupy different social locations, often develop epistemic privileges over their (...) dominant counterparts, especially with respect to finding lacunae in dominant narratives (cf. Hartsock 1987, Collins 1990, Harding 1987 and 1993, and Intemann 2016). This chapter proposes that Diotima’s unique social location – she is an older, non-Athenian woman, well versed in gestation, childbirth, and childrearing – disposes her to know something about erōs that the dominant group, the Athenian pederasts, do not. What she knows is that having erōs for something does not merely inspire one to pursue or possess it; by attending to how erōs affects women, she argues that it also manifests as a desire to reproduce what is beloved and care for it. Diotima’s standpoint is epistemically revelatory, then, because it establishes that love is far more than the pursuit of beauty; it is a creative and laborious act. Moreover, her standpoint is ethically revelatory, since she argues that the highest form of love entails fostering and nourishing virtue in others. Understood in this way, Diotima is espousing something like a protofeminist ethics of care. (shrink)

What are the ethical responsibilities of the historian in an age of mass murder and hyperreality? Can one be postmodern and still write history? For whom should history be written? Edith Wyschogrod animates such questions through the passionate figure of the "heterological historian." Realizing the philosophical impossibility of ever recovering "what really happened," this historian nevertheless acknowledges a moral imperative to speak for those who have been rendered voiceless, to give countenance to those who have become faceless, and hope (...) to the desolate. Wyschogrod also weighs the impact of modern archival methods, such as photographs, film, and the Internet, which bring with them new constraints on the writing of history and which mandate a new vision of community. Drawing on the works of continental philosophers, historiographers, cognitive scientists, and filmmakers, Wyschogrod creates a powerful new framework for the understanding of history and the ethical duties of the historian. BackgroundInformed consent as stipulated in regulatory human research guidelines requires volunteers to be well-informed about what will happen to them in a trial. However, researchers may be faced with the challenge of how to ensure that a volunteer agreeing to take part in a clinical trial is truly informed. This study aimed to find out volunteers’ comprehension of informed consent and voluntary participation in Human Immunodeficiency Virus (HIV) clinical trials during the registration cohort.MethodsWe conducted a qualitative study among volunteers who (...) were enrolled in the registration cohort of HIV clinical trials in Dar es Salaam, Tanzania. A purposive sampling strategy was used to obtain twenty study participants. The data were collected between June and September 2020 using a semi-structured interview guide. In-depth interviews were used to collect the data to obtain deep insights of the individual study participants on the comprehension of informed consent and participation in the clinical trial. A thematic analysis approach was used to analyze the data. Themes and subthemes were supported by the quotes from the participants.ResultsVolunteers described comprehension of informed consent from different perspectives. They reported that various components of the informed consent such as study procedure, confidentiality, risk and benefits were grasped during engagement meetings. Furthermore, the volunteers’ decision to participate in the registration cohort was voluntary. However, trial aspects such as health insurance, free condoms, and medical checkups could have indirectly influenced their reluctance to withdraw from the study.ConclusionEngagement meetings may increase the comprehension of informed consent among potential participants for HIV clinical trials. However, trial incentives may influence participation, and thus future research should focus on the challenges of giving incentives in the study. This will ensure comprehension and voluntary participation in the context of HIV clinical trials. (shrink)
