Analysis of sites of newly integrated DNA in cellular genomes is important to several fields, but methods for analyzing and visualizing these datasets are still under development. Here, we describe tools for data analysis and visualization that take as input integration site data from our INSPIIRED pipeline. Paired-end sequencing allows inference of the numbers of transduced cells as well as the distributions of integration sites in target genomes. We present interactive heatmaps that allow comparison of distributions of integration sites to (...) genomic features and that support numerous user-defined statistical tests. To summarize integration site data from human gene therapy samples, we developed a reproducible report format that catalogs sample population structure, longitudinal dynamics, and integration frequency near cancer-associated genes. We also introduce a novel summary statistic, the UC50, which provides a single number summarizing possible clonal expansion. Using these tools, we characterize ongoing longitudinal characterization of a patient from the first trial to treat severe combined immunodeficiency-X1, showing successful reconstitution for 15 years accompanied by persistence of a cell clone with an integration site near the cancer-associated gene CCND2. Especially in the United States, a widespread discussion about its role in moral theology has been initiated, a discussion modeled on Aristotle's Ethics, particularly as Aristotle's thought was developed in the Middle Ages by Thomas Aquinas. Accompanying this rediscovery of virtue is a criticism of modern ethical theories. In view of this ethical pluralism, certain philosophers have undertaken to set up ethical norms through reflection on justice, using the methods of discussion and decision prevalent in our democratic societies. The initiative has been designated as procedural ethics.2 In this rather chaotic situation, other writers have thought a return to Aristotelian ethics opportune-a move that would enable us to reconnect with the long tradition of virtue-based morality represented therein, while bringing it up to date. In fact, the introduction of the concept of virtue offers many opportunities for the shaping of a morality that takes the human person into account. Virtue is a dynamic human quality acquired through education and personal effort. It forms character and assures continuity in action. Furthermore, it is set within the framework of community and a strong tradition, to whose development it contributes. Teaching on virtue would seem to be a good corrective for excessive individualism. This is what the socalled "communitarian" trend has emphasized.3 The debate on virtue has also surfaced within traditional Catholic teaching. Since the first half of this century, under the inspiration of St. Thomas, several authors of moral theology textbooks have undertaken to reorganize special morality on the basis of the virtues rather than the ten commandments, as had been done since the seventeenth century.4 Admittedly, the change Aristotelian construct of the virtues held by Macintyre. There has been plenty of empirical evidence which shows that the single-particle picture holds to a good approximation in atomic nuclei. In this picture, protons and neutrons move independently inside a mean-field potential generated by an interaction among the nucleons. This leads to the concept of nuclear shells, similar to the electronic shells in atoms. In particular, the magic numbers due to closures of the nucleonic shells, corresponding to noble gases in elements, have been known to play an important (...) role in nuclear physics. Here we propose a periodic table for atomic nuclei, in which the elements are arranged according to the known nucleonic shells. The nuclear periodic table clearly indicates that nuclei in the vicinity of the magic numbers can be understood in terms of a shell closure with one or two additional nucleons or nucleon holes, while nuclei far from the magic numbers are characterized by nuclear deformation. In the quest for Buddhist reality, the inevitable comparison is made between it and the Brahmanic concept of supreme reality. In some quarters, it is alleged that both systems point at an identical nature of reality and maintain a similar method in arriving at it. After all, the historical Buddha was a Brahmin oriented in the Upaniṣadic tradition. He also engaged himself in the prevailing disciplinary practice of yoga to overcome the ill‐nature of the ordinary self (ātman) and like other (...) fellow Brahmins, he consummated his goal by immersing himself in the Supreme Self (Ātman/Brahman). Proponents go even further to identify this Supreme Self with the contents of Buddhist nirvāna and conclude rather facilely that Buddhism is nothing but an extension of Brahmanic thought. (shrink)

Samuel Johnson has an interesting comment on consequences and the telling of “white lies.” For example “Sick People and Children are often to be deceived for their Good.” David Hume apparently endorses this concept in one of his letters. Both Johnson and Rousseau anticipate Kant’s argument about consequences in that one is to tell the truth under all circumstances. Hume, I argue, would take issue with this claim in that there are cases that warrant telling white lies. Elsewhere he speaks (...) about “harmless liars” who indulge in “lying or fiction... in humorous stories.” And he says “Noble pride and spirit may openly display itself when one lies under calamity [defamation or slander] or opposition of any kind,” especially if the opposition puts one’s life in grave danger, so one’s self-preservation is threatened. Under situations like these, lying is justified. In regard to fiction, if lying is for the purpose of entertainment and where “truth is not of any importance,” it is permissible. These cases are discussed in some detail, and they offer, along with their analysis, a pragmatic defense of Hume’s position. (shrink)

Images of Native Americans and of aspects of Native American culture are common in advertisements in the United States. Three such images can be distinguished — the Noble Savage, the Civilizable Savage and the Bloodthirsty Savage images. The aim of this paper is to argue that the use of such images is not morally acceptable because these images depend upon an underlying conception of Native Americans that denies that they are human beings. By so doing, it also denies to (...) them any moral standing and thus any claim to moral consideration and treatment. I begin by arguing that the traits which are distinctively human are fostered only within a cultural framework consisting of the accumulated knowledge and activities of a group of human beings. I then argue that savages are conceptualized as natural and cultureless beings. Furthermore, within the traditional Western conceptualization of the world mere natural objects have no moral standing. Thus, it follows that Native Americans insofar as they are also merely natural objects would have no standing or status as moral beings. The conception of Native Americans as savages undercuts the very conditions for the possibility of moral respect. I then turn to an application of these principles to some current commercial uses of images of Native Americans and other indigenous peoples. (shrink)

The Periodic Table has the column of the noble gas atoms (He, Ne, Ar, Kr, Xe, Rn) as one of its main pillars. Indeed the inert chemical nature of their closed shell structure is so striking that it is sometimes extended to all such structures. Is it true however that any closed shell, say a closed d-subshell will denote a lack of chemical activity? Take the noble metals for instance, so renowned for their catalytic capacity. Platinum has 10 (...) electrons in its valence shell which makes one of its excited states to be a closed 5d10–6s0 state. Surely this state would not be expected to be crucial to the catalytic activity of platinum, or would it? Or take palladium whose ground state is precisely the 4d10–5s0 state, should we expect that an isolated Pd atom at near zero-point temperature would attack a closed-shell hydrogen molecule efficiently? We shall here show that this is precisely the case; the closed-shell excited states of nickel and platinum are indeed crucial, through symmetry avoided crossings, for their reactivity. Other valuable catalysts as ruthenium depend on their excited states with maximal d-shell occupancy for their activity. The most notable confirmation of this new finding; that closed d-shells are vital to the catalytic activity of noble metals however, is the case of palladium whose closed-shell ground state is indeed capable of attacking hydrogen and hydrocarbon molecules even at temperatures well below 10 K as was predicted theoretically and immediately confirmed experimentally. Though widely cited as a critic of the early ethological program presented by Konrad Lorenz and Niko Tinbergen, other significant aspects of Lehrman’s career and research have not received historical attention. In this paper, I offer a fuller account of Lehrman’s work by situating his debate with ethologists within the larger context of Lehrman’s early scholarly development under G. K. Noble and T. C. Schneirla, by (...) examining his scientific research on the ring-dove as well as his epistemological views about the best way to understand animal behaviour, and by presenting his leadership in institution and network-building of interdisciplinary approaches to animal behavior. In the last book of The Consolation of Philosophy, Boethius develops his solution to the problem of divine foreknowledge and free choice. Interpreters standardly hold that this problem and his solution to it presuppose causal indeterminism. In this paper, I argue that Boethius, following a Neoplatonist view found in Proclus, is a causal determinist and compatibilist and maintains that God’s providential knowledge ensures the occurrence of all the events he knows. This alternative interpretation offers a better fit with Boethius’s text (...) and its historical antecedents and provides resources to address the main criticisms of his solution. (shrink)

Ancient Indian philosophies consider self-realization as a fundamental concept and aim of human life, which appears theoretically similar to the self-actualization concept of the West. This article compares and contrasts the self-actualization concept with the views of ancient Indian wisdom to create a model. Both ideas strive for a more elevated Self, unleashing our potential or the realization/actualization of the true Self. From the Indian Vedanta philosophy emerged the Panchakosha theory of personality, which provides a structural framework for human states (...) of consciousness leading to higher development and transcendental identity. According to the scriptures, any form of stress is due to an underdeveloped state of consciousness at these levels or due to disconnection from the true Self; once an individual comes in contact with the ‘true Self’, the problems of life are eliminated. Yoga is a systematic process and way of human development, and applying the principles of Yoga philosophy in life will help in transcending one from the lower Self to the higher Self, awakening human potential, and becoming self-actualized. Yogis, who practice Yoga, could be responsible for a positive shift towards sustainable communities or transformative environments through enhanced mindfulness, self-actualization, and social capital demonstrated by enhanced moral values, trust, engagement, and acceptance. This theoretical model from classical Indian thought has the potential to provide the needed guidelines to create more self-actualized people. The dimension of well-being and prescriptions on how to reach this state of highest well-being is the essence of Indian philosophy. (shrink)

Equality is a concept that is often used in health care discussions about the allocation of resources and the design of health care systems. In secular discussions and debates the concept of equality is highly controverted and can take on many different specifications. One might think that Christians hold a common understanding of equality. A more careful study, though, makes it quite clear that equality is just as controversial among different Christian communities as it is in the secular world.

This essay delineates points of agreement and disagreement between Plato and Nietzsche with respect to the original Heraclitean argument that the underlying dynamic connective structure of the whole is ‘strife’. Also discussed is the issue of how each philosopher understands life itself, as a general process, to be related to the wider processive whole. The paper analyses how the Heraclitean understanding of the natural whole influences each philosopher's interpretation of the political structures of man. The analysis attempts to demonstrate why (...) for Nietzsche, on one hand, the value forms of the traditional polis are more in accord with human nature and nature in toto than is philosophy and why for Plato, on the other hand, when the value forms of the polis are superseded by those of philosophy it is a natural process rather than contrary to nature. Level statistics and nodal point distribution in a rectangular semiconductor quantum dot are studied for different degrees of spin-orbit coupling. The chaotic features occurring from the spin-orbit coupling have no classical counterpart. Using experimental values for GaSb/InAs/GaSb semiconductor quantum wells we find that level repulsion can lead to the semi-Poisson distribution for nearest level separations. Nodal lines and nodal points are also investigated. Comparison is made with nodal point distributions for fully chaotic states.
