In vitro reconstitution assays are commonly used to study biological membrane fusion. However, to date, most ensemble and single‐vesicle experiments involving SNARE proteins have been performed only with lipid‐mixing, but not content‐mixing indicators. Through simultaneous detection of lipid and small content‐mixing indicators, we found that lipid mixing often occurs seconds prior to content mixing, or without any content mixing at all, during a 50‐seconds observation period, for Ca2+‐triggered fusion with SNAREs, full‐length synaptotagmin‐1, and complexin. Our results illustrate the caveats of (...) commonly used bulk lipid‐mixing fusion experiments. We recommend that proteoliposome fusion experiments should always employ content‐mixing indicators in addition to, or in place of, lipid‐mixing indicators. (shrink)

Poverty alleviation consumption, which we call altruistic consumption, has become a new effective way to help vulnerable groups, but there are a few empirical researches on poverty alleviation through consumption. This article takes China's poverty alleviation actions as the research object, investigates and studies the relationship between altruistic consumption motivations and altruistic consumption behaviors that aim for vulnerable groups. It is found that altruistic consumption behavior is mainly affected by benefit group motivation, benefit morality motivation, benefit demander motivation, and benefit (...) supplier motivation. There is a correspondence between the four altruistic consumption motivations and the four altruistic consumption behaviors. The strength of altruistic consumption motivations changes with changes in altruistic buying behavior. The strength of benefit group motivation decreases with the increase in the times of altruistic purchases, while the strength of benefit morality motivation, benefit demander motivation, and benefit supplier motivation increase with the increase in the times of altruistic purchases. Among the four kinds of altruistic consumption motivations that affect the times of altruistic purchases, the benefit demander motivation has a relatively greater influence. The results of this study have important guiding significance for vulnerable groups to formulating targeted proactive marketing strategies, preventing the altruistic consumption relationship dissolution, and realizing sustainable altruistic consumption. The utilisation of human organoids has the potential to expedite the cycle of biological innovation significantly, yet it also raises a series of ethical and societal concerns. This study aims to evaluate the perceptions and attitudes of researchers regarding the ethical and social challenges associated with the research and application of human organoids. A 20-item questionnaire was developed to assess various aspects: four items evaluated the overall understanding of organoids, 13 items addressed ontological, ownership, informed consent and additional ethical issues, (...) while the remaining four items focused on ethical governance and regulatory concerns. This questionnaire was distributed to attendees of the 3rd China Symposium on 3D Cell Culture and Organoids in May 2023. Basic descriptive statistical analyses and chi-square tests were conducted. The work has been reported in line with the ARRIVE guidelines 2.0. A substantial majority (over 70%) of participants expressed concerns that the research and application of human organoids might raise ethical issues, such as moral status, informed consent and commercialisation. In contrast, a minor segment (4.5%, n = 8) reported feelings of disgust or nausea towards human-animal neural chimeras, with a significant gender difference (p = 0.004). Regarding informed consent for human organoid samples, over 90% of researchers rejected blanket consent for all purposes, and more than half preferred a model of continuous consent, which entails resoliciting consent for new uses. Further, over 40% of participants believed that the commercialisation of human organoids is dependent on their specific type. Our findings show that some researchers held that as long as human embryonic stem cells are not utilised in human organoid research and application, there are no ethical relevant issues. Most researchers also showed indifference towards the concern of moral status and humanisation of animals. The majority believe that the “consent for governance” model is preferable than broad consent or specific consent. We believe to realise the promise of human organoids for medicine, navigating complex ethics tensions and proactively engaging diverse stakeholders are required. Only with prudent, thoughtful and effective guidance, mutual understanding and trust between scientists and public can be fostered; the promising field of organoids can progress rapidly and responsibly. In January 2019, What is Peppa?, a promotional video for a Chinese New Year movie, received widespread attention in China immediately after its release. This paper explains the success of the video from the perspective of visual framing, and employs Systemic Visual Grammar to analyze how visual semiotics are applied to frame prominent topics so as to achieve promotional purposes. The results show that: (i) visual semiotics are used to frame the topic of empty nesters, implying that this issue brings (...) negative consequences and evoking the audience’s empathy and concern; (ii) visuals frame the topic of family love, in which the main protagonist’s actions are highlighted; and (iii) the topic of family reunion is mainly framed and realized by delicate compositional arrangements. Using various visual semiotics at the denotative, stylistic-semantic, connotative, and ideological levels, this video establishes both visual and emotional communication between the main character and the audience, making the promotional purpose more successful. The results highlight and specify the important roles of visual frames in promotional videos, and the integration of Peppa with traditional Chinese culture, love, and family reunion represents a visual approach to glocalization. Teaching research activities in China are practical and reflective research of teachers on teaching. These required activities are meant to ensure quality education and facilitate the professional development of teachers. However, in TRA, teachers encounter many challenges such as low efficiency and weak team collaboration. These problems make it hard to achieve the expected outcomes. W Primary School reformed its activities using Change Laboratory, a formative intervention approach to workplace learning and development based on activity theory. The data collected included (...) seven recorded meetings in the Change Laboratory. The conversations in the meetings were then transcribed into texts. A deductive method of content analysis was used to code the data, focusing on categorizing comments of teachers about the transformative agency. The findings showed the following: There were five types of transformative agencies, namely, resisting, criticizing, explicating, envisioning, and committing to actions. Resisting and criticizing were represented less frequently, and taking action did not emerge as a type of transformative agency. The comments about transformative agency about tools were more frequent than comments about other elements in the activity system. There were some differences in the expression of transformative agency across participants. At the end of this study, the implications for the development of TRA are discussed. (shrink)

Bioethical debates on the use of human embryos and oocytes for stem cell research have often been criticized for the lack of empirical insights into the perceptions and experiences of the women and couples who are asked to donate these tissues in the IVF clinic. Empirical studies that have investigated the attitudes of IVF patients and citizens on the donation of their embryos and oocytes have been scarce and have focused predominantly on the situation in Europe and Australia. This article (...) examines the viewpoints on the donation of embryos for stem cell research among IVF patients and students in China. Research into the perceptions of patients is based on in-depth interviews with IVF patients and IVF clinicians. Research into the attitudes of students is based on a quantitative survey study. The empirical findings in this paper indicate that perceptions of the donation of human embryos for stem cell research in China are far more diverse and complex than has commonly been suggested. Claims that ethical concerns regarding the donation and use of embryos and oocytes for stem cell research are typical for Western societies but absent in China cannot be upheld. The article shows that research into the situated perceptions and cultural specificities of human tissue donation can play a crucial role in the deconstruction of politicized bioethical argumentation and the assumptions about “others” that underlie socio-ethical debates on the moral dilemmas of technology developments in the life sciences. With the development of China’s higher education, China has become increasingly popular as an education destination for international students. To explore the rationale and the influencing factors for them to attend Chinese universities, this research adopted a questionnaire survey and interviews with international students from a top university in China. The data were analyzed from four aspects: home countries’ push factors, China’s pull factors, university attractiveness factors and personal choice factors. The findings show that university attractiveness factors are more significant (...) than the China’s pull factors and the home countries push factors; China’s pull factors have significant influence on the university attractiveness factors; For students from different regions, the factors of China’s pull, home country’s push and university attractiveness play significantly different role in affecting their choice of university, with Asian students much more affected than those from other parts of the world. Besides, individual subjective norms and individual value choices also affect students’ choices to study abroad. (shrink)

We here review primary methods used in quantifying and mapping 5‐hydroxymethylcytosine (5hmC), including global quantification, restriction enzyme‐based detection, and methods involving DNA‐enrichment strategies and the genome‐wide sequencing of 5hmC. As discovered in the mammalian genome in 2009, 5hmC, oxidized from 5‐methylcytosine (5mC) by ten‐eleven translocation (TET) dioxygenases, is increasingly being recognized as a biomarker in biological processes from development to pathogenesis, as its various detection methods have shown. ObjectiveThis study aimed to examine the effectiveness and safety of the Brain-computer interface in treatment of upper limb dysfunction after stroke.MethodsEnglish and Chinese electronic databases were searched up to July 2021. Randomized controlled trials were eligible. The methodological quality was assessed using Cochrane’s risk-of-bias tool. Meta-analysis was performed using RevMan 5.4.ResultsA total of 488 patients from 16 RCTs were included. The results showed that the meta-analysis of BCI-combined treatment on the improvement of the upper limb function showed statistical significance [standardized (...) mean difference : 0.53, 95% CI: 0.26–0.80, P < 0.05]; BCI treatment can improve the abilities of daily living of patients after stroke, and the analysis results are statistically significant ; and the BCI-combined therapy was not statistically significant for the analysis of the Modified Ashworth Scale.ConclusionThe meta-analysis indicates that the BCI therapy or BCI combined with other therapies such as conventional rehabilitation training and motor imagery training can improve upper limb dysfunction after stroke and enhance the quality of daily life. Aerial work platform is a kind of engineering vehicle which is used for hoisting personnel to the appointed place for maintenance or installation. Based on the dynamics model considering the flexible deformation existing in the arm system of folding-boom aerial platform vehicle, this study presents a NN-based backstepping controller used for trajectory tracking control of work platform. The proposed controller can reduce tracking error of work platform and suppress the vibration simultaneously by using the RBF neural network system to compensate (...) model uncertainties and disturbances. Furthermore, we prove that the whole system is stable and convergent by Lyapunov stability theorem. In addition, we give the simulation results which show that the good control performance of the designed controller for trajectory tracking and vibration inhabiting of work platform in the case of model uncertainties. Collaborative research practices in the field of plant genetic resources must follow the principles of fairness and equity as defined in the Convention on Biological Diversity (CBD) and in the International Treaty on Plant Genetic Resources for Food and Agriculture (ITPGRFA). In this context the concepts of fairness and equity generally refer to the substantive and procedural dimensions associated with sharing the benefits of this research. But neither term is clearly defined by these international treaties, and the meanings attributed to (...) the concepts vary among different societies. This paper looks at the question of how to account for the diversity among value systems when conducting research that implicates diverse stakeholders and respects the requirements of fairness and equity. We incorporated an auto-ethnography method developed as part of a multi-stakeholder network involved in research projects on plant genetic resources in West Africa. A theatrical device was used as a framework for testing the principles of fairness and equity, helping us to collectively identify feelings of injustice, and explore the conditions for making collaborative research practices more ethical in ways that respect the perspectives of different stakeholders. In an environment of extensive political and socio-cognitive inequality, this approach makes it possible to explain the criteria relating to interactional justice and expectations in terms of socio-political and socio-emotional benefits. It also invites us to consider the principles of fairness and equity in a framework of ethical competence that goes beyond international directives. (shrink)

Resource saving has become an integral aspect of manufacturing in industry 4.0. This paper proposes a multisystem optimization algorithm, inspired by implicit parallelism of heuristic methods, to solve an integrated production scheduling with resource saving problem in textile printing and dyeing. First, a real-world integrated production scheduling with resource saving is formulated as a multisystem optimization problem. Then, the MSO algorithm is proposed to solve multisystem optimization problems that consist of several coupled subsystems, and each of the subsystems may contain (...) multiple objectives and multiple constraints. The proposed MSO algorithm is composed of within-subsystem evolution and cross-subsystem migration operators, and the former is to optimize each subsystem by excellent evolution operators and the later is to complete information sharing between multiple subsystems, to accelerate the global optimization of the whole system. Performance is tested on a set of multisystem benchmark functions and compared with improved NSGA-II and multiobjective multifactorial evolutionary algorithm. Simulation results show that the MSO algorithm is better than compared algorithms for the benchmark functions studied in this paper. Finally, the MSO algorithm is successfully applied to the proposed integrated production scheduling with resource saving problem, and the results show that MSO is a promising algorithm for the studied problem. BackgroundRepetitive transcranial magnetic stimulation applied to the mylohyoid cortical region has positive clinical effects on post-stroke. Therefore, we conducted a meta-analysis to investigate the efficacy of rTMS for patients with post-stroke dysphagia.MethodsAccording to PRISMA guidelines, we searched the databases of MEDLINE, Cochrane Library, Embase, Web of Science, CNKI, Wangfang. We searched for studies of randomized controlled trials of rTMS to treat dysphagia after stroke and screened by inclusion and exclusion criteria. Features of RCTs were extracted. The heterogeneity of the trials (...) was measured by I2 statistic.ResultsIn total, 11 RCTs with 463 dysphagia patients fulfilled our inclusion criteria. In our analysis, rTMS demonstrated a great beneficial effect for post-stroke dysphagia when combined with traditional swallowing exercises. Moreover, a greatly significant difference was noted based on stimulation frequency. Additionally, no significant difference was observed based on stimulation site.ConclusionsOverall, rTMS can effectively accelerate the improvement of swallowing function in patients with post-stroke swallowing disorders. At present, most of the research in the field of medical-assisted diagnosis is carried out based on image or electronic medical records. Although there is some research foundation, they lack the comprehensive consideration of comprehensive image and text modes. Based on this situation, this article proposes a fusion classification auxiliary diagnosis model based on GoogleNet model and Bi-LSTM model, uses GoogleNet to process brain computed tomographic images of ischemic stroke patients and extract CT image features, uses Bi-LSTM model to extract (...) the electronic medical record text, integrates the two features using the full connection layer network and Softmax classifier, and obtains a method that can assist the diagnosis from two modes. Experiments show that the proposed scheme on average improves 3.05% in accuracy compared to individual image or text modes, and the best performing GoogleNet + Bi-LSTM model achieves 96.61% accuracy; although slightly less in recall, it performs better on F1 values, and has provided feasible new ideas and new methods for research in the field of multi-model medical-assisted diagnosis. Previous studies have explored the impact of the cost ratio of individual solutions versus collective solutions on people’s cooperation tendency in the presence of individual solutions. This study further explored the impact of team credibility on people’s propensity to cooperate in the presence of individual solutions. Study 1 investigated the influence of different level of altruistic tendencies or the self-interest tendencies of teammates on participants’ decision-making. Study 2 explored the influence of the distribution of altruistic tendencies or self-interest tendencies on (...) participants’ decision-making. The results of Study 1 showed that the proportion of participants who chose the collective solution increased with an increase in the altruistic tendencies of the team. When the altruistic tendencies of the teammates reached a certain value, the proportion of participants taking the collective solution showed a trend to stabilize. Furthermore, the proportion of participants who chose the individual solution increased with the increase in the self-interest tendencies of the team. When the self-interest tendencies of the teammates reached a certain value, the individual solution was stably adopted. The results of Study 2 showed that with the total altruistic tendency remaining unchanged, the more altruistic group members that altruistic tendencies were allocated to, the higher a participant’s level of trust in the team would be, which showed the decentralized effect of altruistic tendencies. In the case that the total self-interest tendency was unchanged, the fewer self-interest group members the self-interest tendencies were allocated to, the higher a participant’s level of trust in the team would be, which showed the convergent effect of self-interest tendencies.
