Though hippocampal volume reduction is a pathological hallmark of schizophrenia, the molecular pathway responsible for this degeneration remains unknown. Recent reports have suggested the potential role of impaired blood-brain barrier function in schizophrenia pathogenesis. However, direct evidence demonstrating an impaired BBB function is missing. In this preliminary study, we used immunohistochemistry and serum immunoglobulin G antibodies to investigate the state of BBB function in formalin-fixed postmortem samples from the hippocampus and surrounding temporal cortex of patients with schizophrenia and controls without (...) schizophrenia matched for age, sex, and race. Since a functional BBB prevents the extravasation of IgGs, detection of IgGs in the parenchyma is used as direct evidence of BBB breakdown. We also developed a semi-quantitative approach to quantify the extent of IgG leak and therein BBB breach. Analysis of our immunohistochemistry data demonstrated a significantly higher incidence of IgG leak in patients with schizophrenia compared to controls. Further, BBB permeability was significantly higher in advanced-age patients with schizophrenia than both advanced-age controls and middle-aged patients with schizophrenia. Male patients with schizophrenia also demonstrated a significant increase in IgG permeability compared to control males. Interestingly, the extravasated IgGs also demonstrated selective immunoreactivity for neurons. Based on these observations, we suggest that BBB dysfunction and IgG autoantibodies could be two key missing pathoetiological links underwriting schizophrenia hippocampal damage. (shrink)

Seeking a scientific basis for understanding and treating mental illness, and inspired by the work of Ivan Pavlov, American physiologists, psychiatrists and psychologists in the 1920s turned to nonhuman animals. This paper examines how new constructs such as “experimental neurosis” emerged as tools to enable psychiatric comparison across species. From 1923 to 1962, the Cornell “Behavior Farm” was a leading interdisciplinary research center pioneering novel techniques to experimentally study nonhuman psychopathology. Led by the psychobiologist Howard Liddell, work at the Behavior (...) Farm formed part of an ambitious program to develop new preventative and therapeutic techniques and bring psychiatry into closer relations with physiology and medicine. At the heart of Liddell’s activities were a range of nonhuman animals, including pigs, sheep, goats and dogs, each serving as a proxy for human patients. We examine how Pavlov’s conceptualization of ‘experimental neurosis’ was used by Liddell to facilitate comparison across species and communication between researchers and clinicians. Our close reading of his experimental system demonstrates how unexpected animal behaviors and emotions were transformed into experimental virtues. However, to successfully translate such behaviors from the animal laboratory into the field of human psychopathology, Liddell increasingly reached beyond, and, in effect, redefined, the Pavlovian method to make it compatible and compliant with an ethological approach to the animal laboratory. We show how the resultant Behavior Farm served as a productive “hybrid” place, containing elements of experiment and observation, laboratory and field. It was through the building of close and more naturalistic relationships with animals over extended periods of time, both normal and pathological, and within and outside of the experimental space, that Liddell could understand, manage, and make useful the myriad behavioral complexities that emerged from the life histories of experimental animals, the researchers who worked with them, and their shared relationships to the wider physical and social environments. ABSTRACT:Neuroethics, the study of the cognitive and neural mechanisms underlying ethical decision-making, is a growing field of study. In this review, we identify and discuss four themes emerging from neuroethics research. First, ethical decision-making appears to be distinct from other types of decision-making processes. Second, ethical decision-making entails more than just conscious reasoning. Third, emotion plays a critical role in ethical decision-making, at least under certain circumstances. Lastly, normative approaches to morality have distinct, underlying neural mechanisms. The revolution involving the foundations of the physical sciences heralded by relativity and quantum theories has been stimulating philosophers for many years. Both of these comprehensive sets of concepts have involved profound challenges to traditional theories of epistemology, ontology, and language. This volume gathers six experts in physics, logic and philosophy to discuss developments in space exploration and nuclear science and their impact on the philosophy of science.

Premise: our representational system has had a relatively invariant core throughout human history (cf. Sellars's manifest image). Major theses: (i) When philosophical argument establishes the existence of an entity, that entity is a representing, not a represented. (ii) Most of the documents in the history of philosophy are on a par (as dialogical resources) with current philosophical literature for establishing or controverting such existence claims. (iii) The use of mathematics (initially the mathematized neo-Platonism of classical mechanics) allowed modern physical science (...) to break with the perennial system of representation; in consequence, a portion of the representings of modern physical scientists do not belong to the historically invariant core. This limits the dialogical resources of physical science and the applicability of arguments from perennial philosophy to science. It also explains the relative irrelevance of pre-17th century science to contemporary physical scientists in contrast to the relevance of pre-17th century philosophy to contemporary philosophers. It also supports thesis (iv), that logic (broadly conceived) in central to all serious philosophical enterprises, since logic is the central tool for exhibiting and criticizing the rationale(s) of our representational system(s). Individual Differences in Subjective Experience First-Person Constraints on Theories of Consciousness, Subconsciousness, and Self-Consciousness Robert G. ...

Theoretical explication of a growing body of empirical data on consciousness-related anomalous phenomena is unlikely to be achieved in terms of known physical processes. Rather, it will first be necessary to formulate the basic role of consciousness in the definition of reality before such anomalous experience can adequately be represented. This paper takes the position that reality is constituted only in the interaction of consciousness with its environment, and therefore that any scheme of conceptual organization developed to represent that reality (...) must reflect the processes of consciousness as well as those of its environment. In this spirit, the concepts and formalisms of elementary quantum mechanics, as originally proposed to explain anomalous atomic-scale physical phenomena, are appropriated via metaphor to represent the general characteristics of consciousness interacting with any environment. More specifically, if consciousness is represented by a quantum mechanical wave function, and its environment by an appropriate potential profile, Schrödinger wave mechanics defines eigenfunctions and eigenvalues that can be associated with the cognitive and emotional experiences of that consciousness in that environment. To articulate this metaphor it is necessary to associate certain aspects of the formalism, such as the coordinate system, the quantum numbers, and even the metric itself, with various impressionistic descriptors of consciousness, such as its intensity, perspective, approach/avoidance attitude, balance between cognitive and emotional activity, and receptive/assertive disposition. With these established, a number of the generic features of quantum mechanics, such as the wave/particle duality, and the uncertainty, indistinguishability, and exclusion principles, display metaphoric relevance to familiar individual and collective experiences. Similarly, such traditional quantum theoretic exercises as the central force field and atomic structure, covalent molecular bonds, barrier penetration, and quantum statistical collective behavior become useful analogies for representation of a variety of consciousness experiences, both normal and anomalous, and for the design of experiments to study these systematically. Starting with the Greeks, philosophers have been prone to demand certainty in their subject. As we know, this was not a local demand; the prevailing view was that all knowledge, scientific as well as philosophic, must be certain. The demand for philosophic certainty was thus the result of a more general view about knowledge and, equally important, the conviction that philosophy and science are one or, at least, continuous. Eventually, however, although there was agreement on the ideal, disagreement on virtually (...) everything else of importance bred skepticism about philosophic knowledge itself. The heirs to this skepticism in the present century were the logical positivists. -/- As they saw the matter, philosophy is discontinuous with science and, once shorn of its nonsense, contains only logical analyses or clarifications of meanings and not, as had been thought, claims about real existences. Philosophic certainty was retained in principle, although, given the controversial nature of their other claims, it was not emphasized. Science also came down a notch or two. The certainty of mathematics was reaffirmed, but it too was relegated to the realm of meanings; on the other hand, empirical science, since it rests on observation and experiment, was demoted to the level of high probability. -/- In all, the boldness and rigor of the positivist position are unmatched in the present century. Nevertheless, I think it is mistaken. In the present paper, I will argue that, because of difficulties with verificationism and the analytic-synthetic distinction, there is no reason to think that philosophic statements differ in kind from factual claims. The result, I think, is a partial vindication of the long-standing conception that philosophy is continuous with science. As for the other feature of the traditional view of philosophy, viz. certainty, I shall say little. However, if the account I defend here is correct, it should be clear that certainty in philosophy is unattainable. (shrink)

Culp (1994) provides a defense for a form of experimental reasoning entitled 'robustness'. Her strategy is to examine a recent episode in experimental microbiology--the case of the mistaken discovery of a bacterial organelle called a 'mesosome'--with an eye to showing how experimenters effectively used robust experimental reasoning (or could have used robust reasoning) to refute the existence of the mesosome. My plan is to criticize Culp's assessment of the mesosome episode and to cast doubt on the epistemic significance of robustness. (...) In turn, I present a different account of the experimental reasoning microbiologists used in arriving at the conclusion that mesosomes are artifacts. I call this form of reasoning 'reliable process reasoning', and close the paper with a brief discussion of how experimental microbiologists justify the claim that an experimental process is reliable. In On Certainty Wittgenstein presents an argument against both G. E. Moore and the Cartesian skeptic, exposing both positions as flawed. His main contention is that what "stands fast" for us-certainty-is not subject to doubt, truth, or falsehood. Whatever is subject to these ascriptions is propositional in form and belongs to our language-games. But certitude is not so subject; certitude is principally non-propositional and therefore stands outside the language-game. Action is the locus of certainty, the things about which we are (...) certain constitute the "foundation" upon which all our knowledge turn. What Moore had once called our "obvious truisms" are, under Wittgenstein's direction, recognized first as inherent in our human natural history, and second, as grounded or based in human societies, cultures, forms of life. It is this human-centered kind of foundationalism that distinguishes Wittgenstein from what Moyal-Sharrock characterizes as the super-human kind of traditional, Cartesian foundationalism. Due to the human emphasis, Wittgenstein's new kind of foundationalism is quite different than the traditional Cartesian variety; it makes no guarantees of absoluteness or universality. Instead, Wittgenstein identifies a common groundwork, a "footing" on which our language-games rest. This new kind of foundationalism shows the way our language-games are anchored to the world. Therefore, in On Certainty, I argue that Wittgenstein presents a new kind of foundationalism. (shrink)
