Microbial systems biology has made enormous advances in relating microbial physiology to the underlying biochemistry and molecular biology. By meticulously studying model microorganisms, in particular Escherichia coli and Saccharomyces cerevisiae, increasingly comprehensive computational models predict metabolic fluxes, protein expression, and growth. The modeling rationale is that cells are constrained by a limited pool of resources that they allocate optimally to maximize fitness. As a consequence, the expression of particular proteins is at the expense of others, causing trade‐offs between cellular objectives (...) such as instantaneous growth, stress tolerance, and capacity to adapt to new environments. While current computational models are remarkably predictive for E. coli and S. cerevisiae when grown in laboratory environments, this may not hold for other growth conditions and other microorganisms. In this contribution, we therefore discuss the relationship between the instantaneous growth rate, limited resources, and long‐term fitness. We discuss uses and limitations of current computational models, in particular for rapidly changing and adverse environments, and propose to classify microbial growth strategies based on Grimes's CSR framework. Currently, two frameworks of causal reasoning compete: Whereas dependency theories focus on dependencies between causes and effects, dispositional theories model causation as an interaction between agents and patients endowed with intrinsic dispositions. One important finding providing a bridge between these two frameworks is that failures of causes to generate their effects tend to be differentially attributed to agents and patients regardless of their location on either the cause or the effect side. To model different types of error attribution, we augmented (...) a causal Bayes net model with separate error sources for causes and effects. In several experiments, we tested this new model using the size of Markov violations as the empirical indicator of differential assumptions about the sources of error. As predicted by the model, the size of Markov violations was influenced by the location of the agents and was moderated by the causal structure and the type of causal variables. (shrink)

Objectives Medical futility at the end of life is a growing challenge to medicine. The goals of the authors were to elucidate how clinicians define futility, when they perceive life-sustaining treatment (LST) to be futile, how they communicate this situation and why LST is sometimes continued despite being recognised as futile. Methods The authors reviewed ethics case consultation protocols and conducted semi-structured interviews with 18 physicians and 11 nurses from adult intensive and palliative care units at a tertiary hospital in (...) Germany. The transcripts were subjected to qualitative content analysis. Results Futility was identified in the majority of case consultations. Interviewees associated futility with the failure to achieve goals of care that offer a benefit to the patient's quality of life and are proportionate to the risks, harms and costs. Prototypic examples mentioned are situations of irreversible dependence on LST, advanced metastatic malignancies and extensive brain injury. Participants agreed that futility should be assessed by physicians after consultation with the care team. Intensivists favoured an indirect and stepwise disclosure of the prognosis. Palliative care clinicians focused on a candid and empathetic information strategy. The reasons for continuing futile LST are primarily emotional, such as guilt, grief, fear of legal consequences and concerns about the family's reaction. Other obstacles are organisational routines, insufficient legal and palliative knowledge and treatment requests by patients or families. Conclusion Managing futility could be improved by communication training, knowledge transfer, organisational improvements and emotional and ethical support systems. The authors propose an algorithm for end-of-life decision making focusing on goals of treatment. (shrink)

Mechanistic explanations – especially mechanistic models – are commonly used to describe living beings. But their usage should be scrutinised closely. This text elucidates the motives, conditions and implications of a so called ‘methodical mechanism’. Due to the fact that humans cannot properly explain certain biotic processes, they have to rely on their technical knowledge to explain them. The history of philosophy and scientific thinking is, therefore, intertwined with the history of technology: the way humans interpret and explain natural phenomena (...) is subject to the technological knowledge of a given time. The danger of this mechanistic approach, however, is that it can lead to false ontological assumptions about nature and humankind’s place therein, resulting in a subjugation of natural processes to instrumental reason. Working with Old High German and Old Saxon texts has become rare in German language and literature studies. Nowadays, the brief introductions to historical linguistics and philology do not suffice to enable students to explore the oldest German texts in significant depth. A new tool could at least help move closer to this dept of textual exploration: an on-line publication of all Old High German and Old Saxon texts with a morphological word-by-word annotation. The data has been collected by the (...) research project ‘Old German Reference Corpus’ (‘Referenzkorpus Altdeutsch’ / ReA) and can already be used for complex database queries. Within the framework of the ‘eHumanities Centre for Historical Lexicography’ (eHumanities-Zentrum für Historische Lexikographie / ZHistLex), it has been converted into a clearly arranged format, focusing on the information needed for comprehending the texts: the ‘Lesekorpus Altdeutsch’ (LeA). This tool can of course not replace the work with text editions, translations and commentaries, but will help pave the way from the first insight into the language to a profound understanding. Additionally, a supplemental both-way linking between every word token and the corresponding entry in the on-line dictionaries of historical German is being prepared. (shrink)

In this paper, the results of an empirical analysis of a set of 416 descriptive case studies published by corporate members of the UN Global Compact are presented. Although these cases cannot be viewed as representative of the Compact itself or of corporate social responsibility (CSR) and development in general, they can illustrate which kinds of projects are deemed appropriate as best practice examples among Compact members, and therefore indicate the direction, in which predominantly voluntary and business‐led CSR might at (...) best be evolving. To help contextualize the analysis, the paper starts with a brief overview of recent academic work on the strengths and limitations of CSR in the light of international development, followed by the empirical analysis of Compact case studies. The results raise doubts regarding the general suitability of contemporary CSR initiatives to tackle some of the most pressing developmental challenges. Instead, only certain topics are commonly addressed, while a number of issues such as anti‐corruption measures or labour rights are underrepresented in the case study sample. Regarding the target regions of the best practice examples, the majority is reported on activities based in OECD countries and a small number of emerging markets such as South Africa, India or China, while neglecting other regions such as sub‐Saharan Africa (excluding South Africa). From a European Union policy perspective, these results indicate that there is a role to play for the state in order to create a better fit between CSR agendas and the actual developmental needs in the South. This chapter provides an account of the Kantian theory of value, showing how a plurality of incommensurable values, namely the fundamentally heterogeneous values of morality and prudence, can be integrated into a complete ordering by appealing to the conditionality of the value of happiness, which allows us to explain how the claims of prudence can be silenced by the claims of morality, thereby solving the Sidgwickian problem of the dualism of practical reason. Moreover, it establishes that the Kantian understanding of (...) silencing is the only way of rendering dualism coherent, given that the question as to what one ought to do, considering all the normative demands to which one is subject, requires either that these demands be commensurable, which presupposes monism, or that these demands never conflict, which can only be ensured in principle by means of conditional value structures. This response article, however, focuses on clarification of conceptual queries, thereby outlining the proposed theory of hallucinations more sharply, discussing its relationship with schizophrenia, and explaining why underconstrained thalamocortical activation may well be a candidate mechanism responsible for acute schizophrenic symptoms other than hallucinations.

This paper contains a development of the consequences of a form of skepticism closely akin to traditional fallibilism. It is contended that fallibilism properly understood is compatible with actual possession of knowledge and with rationally continuing claims to such possession. In order to justify this contention, the notion of a K—game as a species of game played in accordance with rules is developed, first leaning on Brians Skyrms' notion of a rational dialectic and then on independent grounds. The notion of (...) knowledge which emerges from this is compatible with some but not all of the analyses of knowledge which have recently been put forward. (shrink)

After the coup d’état in 2012, the Malian state experienced progressive decomposition in terms of loss of territorial integrity as well as in the functioning of its institutions. Against this backdrop, this article examines life histories of a former student generation whose members were actively involved in the protest movement of 1980. At the time they fought for student rights and democratization under a military junta, but were not able to induce the desired change. Today they portray themselves as committed (...) patriots for whom the state serves as an ongoing point of reference in their identity constructions. Bringing together higher education, protest movement and state decline, I argue that their narratives not only reveal how damaged identities and political visions are narratively repaired, but also, on a symbolic level, their damaged country as well. Gegenstand des Bandes ist eine Profilierung der Kulturphilosophie mit dem Ziel ihrer notigen Selbstbehauptung gegenuber einem gegenwartigen Naturalismus. Ein zentrales Ratsel der Gegenwartsphilosophie ist das Verhaltnis unseres Bewusstseins, der Perspektive der ersten Person machtig und Urheber der Kultur zu sein, zu den Erkenntnissen der Kognitions- sowie der Evolutionswissenschaften seit Darwin. In diesen scheint von der Natur her auf die Kultur ein neues Licht zu fallen. Dies ist ebenso faszinierend wie interessant. Es ist aber auch eine Herausforderung: Der Begriff der "Welt" (...) wird naturwissenschaftlich praokkupiert, Extrapolationen ins Ontologische wie die "kausale Geschlossenheit der Welt" breiten sich auf Kosten fruherer, starker zuruckgenommener Erkenntnis- und Wissenschaftstheorien aus. Wie steht es demgegenuber mit den Beitragen von Kulturphilosophie und Kulturwissenschaften / Geisteswissenschaften? Deutlich wird in den Beitragen im Anschluss an Dilthey, Cassirer, Simmel und weitere Autoren: Die Notwendigkeit und der Eigencharakter kultureller Arbeit mussen offentlich wie in den Wissenschaften wieder bewusst werden.
