Fischer’s approach for structure elucidation of linear aldohexoses is still the most widespread alternative in textbooks for carbohydrates. However, in post-Fischer era, a series of remarkable discoveries and inventions were made in different laboratories, and by their use a more comprehensive and coherent strategy for structure elucidation of linear isomeric aldohexoses can be elaborated. Fischer used the exceptional properties of d-mannose for the knowledge of configuration of C-2 and called it the key of the gate to stereochemistry. We bring the (...) evidence that Fischer had an alternative to this monosaccharide, d-idose. Fischer discovered the correct structure of the enantiomers of tartaric acid by pure intuition but Bijvoet proved it experimentally by means of X-rays. Hybridization, a concept taken by L. Pauling from biology, reconciled physics and chemistry concerning the valence of carbon atom and explained the stereochemical picture that had been empirically elaborated by Van’t Hoff and Le Bel. Other chemical transformations of monosaccharides, useful in the elucidation of their structure, have been also discovered or invented in post-Fischer era. (shrink)

This paper argues that the allure of conspiracy theories lies in their evolutionary origins, specifically in our capacity to communicate unrepresented threats. Drawing on threat-detection psychology and error management theory, it posits that these theories serve as adaptive responses to perceived threats and social coalition-building, rather than as flaws in reasoning.

Especially in functional-typological linguistics, semantic roles have been studied thoroughly, because they constitute a good starting point for any study on argument marking due to their semantically defined nature. However, the very concept of semantic roles is far from being without problems, and there is still no consensus on how the roles are best defined. In this volume, the notion will be discussed from novel perspectives with the aim of providing new insights into our understanding of semantic roles. Two of (...) the papers deal with semantic role clusters, one with semantic roles in verbless constructions, one with diachrony of semantic roles and two with individual semantic roles that have not been studied in too much detail in previous studies. The book may not offer answers to all questions the readers may have, but at least it raises interesting further questions relevant to arriving at a better understanding of semantic roles. In the paper we investigate differences in the skills mismatch in the new and old EU member states. We look at the influence of their different socio-economic characteristics, in particular age and educational attainment, on the level of skills mismatch and link these to the patterns of economic development, in particular the economic transition in Central and Eastern Europe. The results of our analysis show that workers in the NMS compared to the EU-15 have a higher risk of underskilling and (...) a lower risk of overskilling in all age groups. The differences between the two sets of countries are larger for older workers and smaller for younger ones. This indicates that there may be a gradual convergence of the skills match patterns on the European labour market for younger generations and educational levels. (shrink)

Neuroenhancement involves the use of neurotechnologies to improve cognitive, affective or behavioural functioning, where these are not judged to be clinically impaired. Questions about enhancement have become one of the key topics of neuroethics over the past decade. The current study draws on in-depth public engagement activities in ten European countries giving a bottom-up perspective on the ethics and desirability of enhancement. This informed the design of an online contrastive vignette experiment that was administered to representative samples of 1000 respondents (...) in the ten countries and the United States. The experiment investigated how the gender of the protagonist, his or her level of performance, the efficacy of the enhancer and the mode of enhancement affected support for neuroenhancement in both educational and employment contexts. Of these, higher efficacy and lower performance were found to increase willingness to support enhancement. A series of commonly articulated claims about the individual and societal dimensions of neuroenhancement were derived from the public engagement activities. Underlying these claims, multivariate analysis identified two social values. The Societal/protective highlights counter normative consequences and opposes the use enhancers. The Individual/proactionary highlights opportunities and supports use. For most respondents these values are not mutually exclusive. This suggests that for many neuroenhancement is viewed simultaneously as a source of both promise and concern. (shrink)

In a thought experiment we want to test how the emergence of adult neural stem cells could constitute an example for a scientific revolution in the sense of Thomas Kuhn. In his major work, The structure of scientific revolutions, 3rd edn, University of Chicago Press, Chicago (Kuhn 1996), the philosopher of science, Thomas Kuhn, states that scientific progress is not a cumulative process, but new theories appear by a rather revolutionary sequence of events. Kuhn built his theory on landmark events (...) taken from chemistry and physics, lacking examples from biology. Beginning with Ramon y Cajal’s famous quote, no new neurons after birth , from the early years of the twentieth century, and Reynolds and Weiss’s conflicting finding in 1992 of adult neural stem cells giving rise to new neurons, we will test how the finding of neural stem cells in the adult brain matches with Kuhn’s theory. The pivotal problem of defining a paradigm will be our main focus, since the emergence of adult neural stem cells has been acclaimed by the scientific community as the rebuttal of Ramon y Cajal’s paradigm. The pathogenic, Gram‐negative bacteria, Neisseria gon‐orrhoeae, Neisseria meningitidis and Haemophilus influenzae, secrete immunoglobulin A1 proteases into their extracellular surroundings. An extraordinary feature in the secretory pathway of these putative virulence factors is a self‐directed outer membrane transport step allowing the proteins to be secreted autonomously, even from foreign Gram‐negative host cells like Escherichia coli. Here we summarize recent achievements in the understanding of IgA protease outer membrane translocation.

The last decade has seen an explosion of research on the gut microbiota—the trillions of microorganisms that colonize the human gut. It is now clear that interindividual diversity in microbiota composition plays an important role in determining susceptibility to a wide variety of diseases. However, identifying the precise changes in microbiota composition that play causal roles has remained a largely unrealized goal. Here, we propose that functional classifications of microbes based on their interactions with and effects on the host—particularly the (...) host immune system—will illuminate the role of the microbiota in shaping human physiology. We outline the benefits of “functional” classification compared to phylogenetic classifications, and review current efforts at functional classification of the microbiota. Finally, we outline a theoretical framework for classifying host-microbiota interactions. Future advances enabling broader functional classifications of the microbiota promise to revolutionize our understanding of the role of gut microbes in health and disease. Functional classifications of the human microbiota reveal previously hidden patterns and will be key to cracking the microbiota composition code. In March 2022 the US Food and Drug Administration (FDA) published a risk assessment of a recent animal gene editing proposal submitted by Acceligen™. The proposal concerned the possibility of changing the cattle genome to obtain a slicker, shorter hair coat. Using CRISPR-Cas9 it was possible to introduce an intentional genomic alteration (IGA) to the prolactin receptor gene (PRLR), thereby producing PRLR-SLICK cattle. The goal was to diminish heat stress in the cattle by enhancing their heat-tolerance. With regard to unintended (...) alterations (i.e., off-target effects), the FDA stated that the IGA posed a low, but still present, risk to animal safety. The aim of this article is to present some initial insights into the welfare issues raised by PRLR-SLICK cattle by addressing the question: Do SLICK cattle have better welfare than non-SLICK cattle when exposed to heat stress? Two potential welfare concerns are examined. The first is pleiotropy, an issue that arises when one gene affects multiple traits. Given the pleiotropic nature of prolactin, it has been suggested that the IGA for SLICK cattle may also affect their hepatic and other functions. The second concern relates not primarily to direct effects on cattle health, but rather to the indirect risk that this more heat-tolerant animal would just be used in the livestock sector under farming conditions that are such that the net welfare improvement would be non-existent. In this article, an in-depth study and analysis of economic structure are carried out using a neural network fusion release algorithm. The method system defines the weight space and structure space of neural networks from the perspective of optimization theory, proposes a bionic optimization algorithm under the weight space and structure space, and establishes a neuroevolutionary method with shallow neural network and deep neural network as the research objects. In the shallow neuroevolutionary, the improved genetic algorithm based on elite heuristic (...) operation and migration strategy and the improved coyote optimization algorithm based on adaptive influence weights are proposed, and the shallow neuroevolutionary method based on IGA and the shallow neuroevolutionary method based on ICOA are applied to the weight space of backpropagation neural networks. In deep neuroevolutionary method, the structure space of convolutional neural network is proposed to solve the search space design of neural structure search, and the GA-based deep neuroevolutionary method under the structure space of convolutional neural network is proposed to solve the problem that numerous hyperparameters and network structure parameters can produce explosive combinations when designing deep learning models. The neural network fusion bionic algorithm used has the application value of exploring the spatial structure and dynamics of the socioeconomic system, improving the perception of the socioeconomic situation, and understanding the development law of society, etc. The idea is also verifiable through the present computer technology. The determination of amino acid sequences representing the cell‐surface receptors for transferrin,1 asialoglycoprotein,2 polymeric immunoglobulin (IgA/IgM),3 epidermal growth factor (EGF),4 lowdensity lipoprotein (LDL)5 and insulin6 has produced new paradingms for receptor architecture. This review examines common features of the protiens and describes the intriguing functional and evolutionary puzzles that have arisen from them.

Emergence of the novel pathogenic coronavirus SARS‐CoV‐2 and its rapid pandemic spread presents challenges that demand immediate attention. Here, we describe the development of a semi‐quantitative high‐content microscopy‐based assay for detection of three major classes (IgG, IgA, and IgM) of SARS‐CoV‐2 specific antibodies in human samples. The possibility to detect antibodies against the entire viral proteome together with a robust semi‐automated image analysis workflow resulted in specific, sensitive and unbiased assay that complements the portfolio of SARS‐CoV‐2 serological assays. Sensitive, specific (...) and quantitative serological assays are urgently needed for a better understanding of humoral immune response against the virus as a basis for developing public health strategies to control viral spread. The procedure described here has been used for clinical studies and provides a general framework for the application of quantitative high‐throughput microscopy to rapidly develop serological assays for emerging virus infections. Petersen (bio)Keywordsaging, Alzheimer’s disease, dementia, mild cognitive impairment, pharmaceutical industryGraham and Ritchie (2006) have contributed a scholarly document that implores us to reexamine nosological categories and certain diagnostic outcomes. They have chosen mild cognitive impairment (MCI) as the target of their scrutiny and have raised several interesting issues. I would like to comment on their approach and suggest that MCI is a useful (...) clinical entity that does serve a practical function and hopefully will lead to a better quality of life for aging persons.There are several clarifications concerning the construct of MCI that need to be emphasized. Graham and Ritchie have asserted that all persons with MCI eventually evolve to Alzheimer's disease (AD) and have claimed that this is the inevitable outcome of the disorder. Although it is true that many of the early studies on MCI focused on the amnestic subtype as a precursor to AD (Bennett et al. 2002; Ganguli, Dodge, Shen, and DeKosky 2004; Lopez et al. 2003; Petersen et al. 1999), subsequent work has expanded the construct to include prodromal forms of other disorders (Petersen 2004; Winblad et al. 2004). As such, the construct of MCI has become flexible in recent years to account for alternative types of intermediate cognitive impairment.The authors state that MCI addresses those individuals speculated to have an inevitable dementia. Certainly, we do not presume that dementia is inevitable in the aging process; rather, we posit that if certain criteria are fulfilled in persons who are aging, the likelihood of a person progressing to a certain type of dementia is quite high. As such, this is an important precondition about which to learn, because interventional strategies may be available.Graham and Ritchie argue that some cognitive changes with aging are "normal" and trying to classify these individuals as "abnormal" is performing a disservice to much of the aging population. We draw an important distinction between the cognitive changes of normal aging, as they are recognized to exist, and what we feel constitutes the pathologic changes of MCI. We believe that the abnormal cognitive function found in MCI has a high probability of progressing to a greater degree of cognitive dysfunction in a relatively short period of time and these behavioral changes are accompanied by pathologic brain changes that are manifested on magnetic resonance imaging scans, positron emission tomography scans, cerebrospinal fluid biomarkers, and autopsy studies (Hampel et al. 2004; Jack et al. 1999; Markesbery et al. [End Page 45] 2006; Petersen et al. 2006). This type of progression is in contradistinction to other individuals who are experiencing cognitive changes of normal aging. These individuals do not progress rapidly to greater degrees of impairment and autopsy studies reveal that their brains do not harbor the pathologic changes found in the MCI population (Knopman et al. 2003).The cognitive and behavioral changes accompanying persons with MCI are devastating and cannot be ascribed to "senility" or "He is just getting old." It is a mistake to imply that by labeling people with a condition such as MCI we are doing them a disservice. Most of these individuals are seeking medical attention for their perceived difficulties. Consequently, addressing these concerns and educating the individuals on the implications of their symptoms are important services. Although we may not have adequate therapies at present, this does not imply that we should ignore the symptoms.We cannot treat many conditions at present, yet we do not avoid the opportunity to characterize the condition and make a diagnosis. Even if this were part of the aging process, this does not mean that we should ignore the disability and refuse to treat it. Most individuals develop an inflexibility of their optical lenses as they age, yet corrective lenses of one type or another are believed to be extremely beneficial at alleviating this disability associated with the aging phenomenon.
