Enzymes that degrade nucleic acids are emerging as important players in the pathogenesis of inflammatory disease. This is exemplified by the recent identification of four genes that cause the childhood inflammatory disorder, Aicardi‐Goutières syndrome (AGS). This is an autosomal recessive neurological condition whose clinical and immunological features parallel those of congenital viral infection. The four AGS genes encode two nucleases: TREX1 and the hetero‐trimeric Ribonuclease H2 (RNase H2) complex. The biochemical activity of these enzymes was initially characterised 30 years ago (...) but a role in neurological inflammation was entirely unanticipated until they were found to be mutated in AGS. The therapeutic potential of meditation for physical and mental well-being is well documented, however the possibility of adverse effects warrants further discussion of the suitability of any particular meditation practice for every given participant. This concern highlights the need for a personalized approach in the meditation practice adjusted for a concrete individual. This can be done by using an objective screening procedure that detects the weak and strong cognitive skills in brain function, thus helping design a tailored meditation training protocol. (...) Quantitative electroencephalogram (qEEG) is a suitable tool that allows identification of individual neurophysiological types. Using qEEG screening can aid developing a meditation training program that maximizes results and minimizes risk of potential negative effects. This brief theoretical–conceptual review provides a discussion of the problem and presents some illustrative results on the usage of qEEG screening for the guidance of mediation personalization. Progress in emergency and critical care requires that clinical research be performed on patients who are incapable of granting consent for research participation. Analyses of the ethics of such research have left some questions incompletely answered. Why should we be permitted to expose vulnerable patients to research risks without their consent? In particular, how do we justify research interventions that have no potential benefit for participants (nontherapeutic interventions)? This article presents a moral justification for nontherapeutic interventions in emergency research. By (...) relying on a framework for assessing research risks, and by drawing on the example of pediatric research, this justification is founded in how institutional review boards, and society in general, analyze risk. Our justification for emergency research also suggests additional protections for emergency research participants, including a stringent threshold for research risk, that still permit important research to proceed. In the 1890's, Freud attempted to lay out the foundations of a complete, interdisciplinary neuroscience of the mind. The conference that gave rise to this collection of papers, Neuroscience of the Mind on the Centennial of Freud's Project for a Scientific Psychology, celebrated the centrepiece of this work, the famous Project (1895a). Freud never published this work and by 1896 or 1897 he had abandoned the research programme behind it. As he announced in the famous Ch. VII of The Interpretation (...) of Dreams (1900), he would thereafter restrict himself to psychology proper, i.e., what could be done within the ambit of psychological descriptions. The task of characterizing the neural implementation of the psychological was impossible to carry out given the state of knowledge in his time. As Pribram and Gill (1976), Kitcher (1992) and others have demonstrated, Freud's attempt to sketch an interdisciplinary model of the mind using the language of neurons, quantities of energy, etc., was extremely advanced for its time and was probably about as good as could have been done with what was known in 1895. Knowledge of the brain, evolutionary biology, etc., was too limited to allow more. (shrink)

One of the aspects of consciousness deserving of study is what might be called its subjective unity - the way in which, though conscious experience moves from object to object, and can be said to have distinct ‘states', it nevertheless in some sense apparently forms a singular flux divided only by periods of unconsciousness. The work of William James provides a valuable, and rather unique, source of analysis of this feature of consciousness; however, in my opinion, this component of James’ (...) theory of the mind has so far gone under-emphasized in the scholarly literature. This paper undertakes some philosophical geography, trying to draw out and elucidate some of the relevant ideas from James’ corpus, and also subjects those ideas to some analysis to try and assist in judgements of their current importance. This article is devoted to the study of the nature of the influence of elections on the risks of revolutionary destabilization. The authors study different approaches to estimating the probability of revolutionary events in an election year. Different types of revolutionary events are distinguished within the framework of the level of political violence. The primary reasons for the activation of the politically active part of the population, both in autocracies and in transitional political regimes, are identified, including the factionalization of (...) elites, the activation of "sleeping" institutions, the growth of political competence among individuals, the consolidation of the prodemocratic opposition and its institutionalization, electoral fraud and other manifestations of unfair political competition, greater emotional involvement of the population in political processes, and so on. The findings support the conclusion that elections promote peaceful revolutionary episodes and reduce the likelihood of armed uprisings. The novelty of the study lies in the fact that the influence of elections on the risks of revolutionary destabilization is considered in the regime context. The authors explore how elections affect violent or non-violent revolutionary actions in certain political regimes, according to Jack A. Goldstone's typology (full autocracies, partial autocracies, factional democracies, partial democracies and full democracies). The authors conclude that holding elections reduces the risks of armed revolutionary destabilization in all types of regimes, except for factional democracies. On the other hand, unarmed revolutionary action is more likely in an election year - in all regime types except full democracies. At the same time, holding elections primarily increases the risks of unarmed revolutionary uprisings in intermediate regimes / anocracies. However, even among anocracies, factional democracies stand out, where in the election year the risks of unarmed (as well as armed) revolutionary episodes increase significantly more than in partial autocracies and in partial non-factional democracies. At the same time, for consolidated democracies, holding elections is a factor that inhibits rather than provokes revolutionary destabilization. In full autocracies, the risk of unarmed revolutionary action increases in an election year, but not as much as in intermediate regimes. (shrink)

Idiopathic generalized epilepsy (IGE) is a common type of epilepsy and despite an increase in the number of available anti-seizure medications, approximately 20–30% of people with IGE continue to experience seizures despite adequate medication trials. Unlike focal epilepsy, resective surgery is not a viable treatment option for IGE; however, neuromodulation may be an effective surgical treatment for people with IGE. Thalamic stimulation through deep brain stimulation (DBS) and responsive neurostimulation (RNS) have been explored for the treatment of generalized and focal (...) epilepsies. Although the data regarding DBS and RNS in IGE is limited to case reports and case series, the results of the published studies have been promising. The current manuscript will review the published literature of DBS and RNS within the centromedian nucleus of the thalamus for the treatment of IGE, as well as highlight an illustrative case. Programmed cell death (PCD) in unicellular organisms is in some instances an altruistic trait. When the beneficiaries are clones or close kin, kin selection theory may be used to explain the evolution of the trait, and when the trait evolves in groups of distantly related individuals, group or multilevel selection theory is invoked. In mixed microbial communities, the benefits are also available to unrelated taxa. But the evolutionary ecology of PCD in communities is poorly understood. Few hypotheses have been offered (...) concerning the community role of PCD despite its far-reaching effects. The hypothesis we consider here is that PCD is a black queen. The Black Queen Hypothesis (BQH) outlines how public goods arising from a leaky function are exploited by other taxa in the community. Black Queen (BQ) traits are essential for community survival, but only some members bear the cost of possessing them, while others lose the trait In addition, BQ traits have been defined in terms of adaptive gene loss, and it is unknown whether this has occurred for PCD. Our conclusion is that PCD fulfils the two most important criteria of a BQ (leakiness and costliness), but that more empirical data are needed for assessing the remaining two criteria. In addition, we hold that for viewing PCD as a BQ, the original BQH needs to include social traits. Thus, despite some empirical and conceptual shortcomings, the BQH provides a helpful avenue for investigating PCD in microbial communities. (shrink)
