Some 60 years ago chemicals that intercalate between base pairs of duplex DNA were found to amplify frameshift mutagenesis. Surprisingly, the robust induction of frameshifts by intercalators still lacks a mechanistic model, leaving this classic phenomenon annoyingly intractable. A promising idea of asymmetric half‐intercalation‐stabilizing frameshift intermediates during DNA synthesis has never been developed into a model. Instead, researchers of frameshift mutagenesis embraced the powerful slipped‐mispairing concept that unexpectedly struggled with the role of intercalators in frameshifting. It is proposed that (...) the slipped mispairing and the half‐intercalation ideas are two sides of the same coin. Further, existing findings are reviewed to test predictions of the combined “half‐intercalation into the slipped‐mispairing intermediate” model against accumulated knowledge. The existence of potential endogenous intercalators and the phenomenon of “DNA bookmarks” reveal ample possibilities for natural frameshift mutagenisis in the cell. From this alarming perspective, it is discussed how the cell could prevent genome deterioration from frameshift mutagenesis. (shrink)

: In the face of disunification and incommensurability, how can the scientific community maintain itself and establish commensurability? According to Peter Galison's investigations of twentieth-century microphysics, commensurability is achieved through local coordination even in the absence of global meaning: The "strength and coherence" of science is due to diverse, yet coordinated action in trading zones between theorists and experimenters, experimenters and instrument builders, etc. Galison's claim is confronted with Georg Christoph Lichtenberg's establishment of commensurability between unitarians and dualists in the (...) eighteenth century dispute about electrical fluids. The contrast of cases suggests an alternative account: Commensurability may be established through the global coordination of local meaning. And where Galison reifies the disunification of science, this account suggests a dynamic interplay between de facto disunification and an intended unity. This interplay is manifested in the pervasive and ongoing practical concern for the conditions of successful communication in a science that is constantly in-the-making. (shrink)

If all dependent expressions were adjacent some variety of immediate constituent analysis would suffice for grammar, but syntactic and semantic mismatches are characteristic of natural language; indeed this is a, or the, central problem in grammar. Logical categorial grammar reduces grammar to logic: an expression is well-formed if and only if an associated sequent is a theorem of a categorial logic. The paradigmatic categorial logic is the Lambek calculus, but being a logic of concatenation the Lambek calculus can only capture (...) discontinuous dependencies when they are peripheral. In this paper we present the displacement calculus, which is a logic of intercalation as well as concatenation and which subsumes the Lambek calculus. On the empirical side, we apply the new calculus to discontinuous idioms, quantification, VP ellipsis, medial extraction, pied-piping, appositive relativisation, parentheticals, gapping, comparative subdeletion, cross-serial dependencies, reflexivization, anaphora, dative alternation, and particle shift. Natural deduction (for short: nd-) calculi have not been used systematically as a basis for automated theorem proving in classical logic. To remove objective obstacles to their use we describe (1) a method that allows to give semantic proofs of normal form theorems for nd-calculi and (2) a framework that allows to search directly for normal nd-proofs. Thus, one can try to answer the question: How do we bridge the gap between claims and assumptions in heuristically motivated ways? This informal (...) question motivates the formulation of intercalation calculi. Ic-calculi are the technical underpinnings for (1) and (2), and our paper focuses on their detailed presentation and meta-mathematical investigation in the case of classical predicate logic. As a central theme emerges the connection between restricted forms of nd-proofs and (strategies for) proof search: normal forms are not obtained by removing local "detours", but rather by constructing proofs that directly reflect proof-strategic considerations. This paper is the second part of an investigation into Babylonian non-mathematical astronomical texts and the relationships between Babylonian observational and predicted astronomical data. Part I (Gray and Steele 2008) showed that the predictions found in the Almanacs and Normal Star Almanacs were almost certainly made by applying Goal-Year periods to observations recorded in the Goal-Year Texts. The paper showed that the differences in dates of records between the Goal-Year Texts and the Almanacs or Normal Star Almanacs were consistent with (...) the date corrections of a few days which, according to theoretical calculations, should be added to allow for the inexactness of Goal-Year periods. The current paper follows on from our earlier study to consider the effect of the Babylonian calendar on Goal-Year methods of prediction. Due to the fact that the Babylonian calendar year can contain either 12 or 13 months, a Goal-Year period can occasionally be month longer or shorter than usual. This suggests that there should in theory be certain points in the Metonic intercalation cycle where a predicted event occurs one Babylonian month earlier or later than the corresponding event a Goal-Year period later. By comparing dates of lunar and planetary records in the Astronomical Diaries, Goal-Year Texts, Almanacs and Normal Star Almanacs, we show that these month differences between the observational records and the predictions occur in the expected years. This lends further support to the theory that the Almanacs’ and Normal Star Almanacs’ predictions originated from records in the Goal-Year Texts, and clarifies how the Goal-Year periods were used in practice. Today, scholars employ the label ‘narrative Christology’ with relative frequency, though they mean different things when they do so. In this article, I argue that to date, narrative Christology has not yet fully explored the parameters of what it means to attend closely to the narrative form of the Gospels’ presentations of Jesus. I propose, further, that recent developments in literary theory’s so-called ‘New Formalism’ offer useful tools and concepts for moving in that direction. The first part of the article (...) briefly outlines previous scholarship, identifying similarities and differences between various approaches labelled ‘narrative Christology’. The second section introduces the major concepts of New Formalism and how they might extend narrative Christology’s capacity to take narrative form seriously as an object of analysis. The third section of the article offers a case study of a passage that appears in the triple tradition – the intercalated healing stories of Jairus’ daughter and the haemorrhaging woman in Mark 5.21–43; Luke 8.40–56; and Matthew 9.18–26 – in order to explore narrative structure on the micro-level. My ultimate goal is to show how New Formalism can contribute to a more robust narrative Christology and, in so doing, advance our understanding of the distinctive ways in which the Synoptic Gospels construct the figure of Jesus. Mais à quoi peut bien encore servir la politique? Jamais la démocratie n'a été aussi solidement installée et, en même temps, jamais elle n'a été autant menacée par le vide, l'impuissance ou l'abus de pouvoir. Le politique a-t-il encore la main? Le rouleau-compresseur des marchés a-t-il eu raison de sa superbe? L'archaïsme des (...) institutions n'a-t-il pas réduit la démocratie française à un théâtre d'ombres? Les multiples filtres qui s'intercalent entre la sphère politique et le peuple ont-ils vidé le fait politique de son sens, laissant derrière lui des résignés et une société en forme de cocotte-minute? Et pourtant, si notre démocratie ne cesse de se dévorer elle-même, elle se régénère toujours. En effet, le politique est là, comme une braise sous les cendres, prêt à renaître, prêt à écrire encore quelques belles pages de l'histoire de France. C'est comme une étincelle à côté d'un baril de poudre, car il suffit de peu. Quand le voisin de classe de votre enfant est arrêté sans ménagement à la sortie de l'école, avec ses parents parce qu'ils sont sans-papier, voilà que vous vous mobilisez dans des comités de soutien, que vous faites le tour des élus, que vous interpellez le préfet et que vous organisez des manifs! Vous, parent d'élève tranquille qui n'avez jamais eu le moindre intérêt pour la cause des sans-papier! Là est le paradoxe du politique, à la fois décevant, incontournable et enthousiasmant quand la cause s'y prête. Alors, dans ce contexte, comment faire pour changer la vie? Comment débusquer le prêt-à penser politique? A qui s'adresse aujourd'hui la politique? Comment réenchanter ceux qui ne le sont plus? Où mener le combat politique qui est aussi un combat culturel? Quelles vieilles certitudes faut-il déboulonner? Qu'est-ce que l'on a encore à se dire dans notre société et comment la faire tenir ensemble? Toutes ces questions sont au coeur de cet antimanuel politique qui entend prendre à contrepied les stratèges cyniques, les barons inamovibles, les communicants qui pensent le politique comme une publicité, les ambitieux qui veulent la place à tous prix, les résignés qui méprisent tout... Et pourtant, la stratégie, la communication ou l'ambition sont nécessaires à tout projet politique. Arnaud Montebourg ne décerne ni les bons ni les mauvais points comme le maître d'école, mais prend une distance critique sur la politique, sublime et misérable, et sur le politique, omniprésent et invisible. Resumo O presente estudo aborda a articulação entre ciência natural e cosmologia em Tomás de Aquino. Para tanto, são investigadas as concepções da noção de cosmo, mediante a questão: o cosmo pode ser o assunto central da ciência natural em Tomás? No intuito de refletir sobre essa questão, são apresentadas três concepções de cosmo. A primeira é a concepção da metafísica, na qual o cosmo é uma hierarquia de entes. A segunda, por sua vez, é a concepção da cosmografia segundo (...) a qual o cosmo é a relação de esferas intercaladas. A terceira, por fim, é a concepção da cosmologia que entende o cosmo como o conjunto dos corpos simples em perene locomoção natural.The present study aims to articulate natural science and cosmology in Thomas Aquinas. Accordingly, I want to investigate the conceptions of cosmos by the following question: can the cosmos be the central topic of natural science in Aquinas? I present three conceptions of cosmos in order to reflect upon this question. The first is the metaphysical one in which the cosmos is a hierarchy of entities. The second is related to cosmography according to which the cosmos is a relation of intercalated spheres. The third, finally, is the cosmological conception that understands the cosmos as the set of simple bodies in perennial natural locomotion. (shrink)

The change in shape of the C. elegans embryo from an ovoid ball of cells into a worm-shaped larva is driven by three events within the cells of the hypodermis (epidermis): (1) intercalation of two rows of dorsal cells, (2) enclosure of the ventral surface by hypodermis, and (3) elongation of the embryo. While the behavior of the hypodermal cells involved in each of these processes differs dramatically, it is clear that F-actin and microtubules have essential functions in each (...) of these processes, whereas contraction of actomyosin structures appears to be involved specifically in elongation. Molecular analysis of these processes is revealing components specific to C. elegans as well as components found in other systems. Since C. elegans hypodermal cells demonstrate dramatically different behaviors during intercalation, enclosure and elongation, the study of cytoskeletal dynamics in these processes may reveal both unique and conserved activities during distinct epithelial morphogenetic movements. This article sets out to substantiate an understanding of the photographic image as a constellation of scaled relations, with a focus on the significance of historically neglected questions of scale in and for the present. It explores two recurrent themes in Walter Benjamin’s writings: his celebrated methodological-epistemological concept of constellation and his less often remarked fascination for relationships of scale, processes of scaling and the scale effects these produce. These are investigated in light of the mutable and composite character of (...) the contemporary photographic image and its unprecedented scale as a mass form. Key phenomena and prevalent conceptions of photography are approached as already structured by intercalated facts, operations and experiences of scale, scaling and scalability. On this basis, a constellation of three historical essays that examine questions of scale in photography is constructed. The guiding aim of this construction is that such a constellation might stand as a pre-history of scale and its significance in and for photography today. (shrink)

He moves toward these goals through a comprehensive reading of Jean-Jacques Rousseau’s “Socratism” intercalated with Jean Laplanche’s psychoanalytic intervention focused on the (...) universality of untranslatable “enigmatic messages” that shape the (un)conscious of each individual. Through this combination of trajectories, Fain hopes both to deconstruct philosophy’s metaphysical origins along the lines of thinkers from Descartes through Badiou, and also return this fundamental question back to the emergence of the wisdom of eudaimonia, the flourishing of happiness, as a libidinal object.When the teaching, historical periodization, and explication of philosophy takes Rousseau under its wing, it is usually as a political theorist, but, as Fain reminds us, Rousseau explicitly asserts that he is “neither a philosopher nor austere,” but instead a “man of paradox,” a “friend of virtue,” or a “friend of the truth” (qtd. in Fain 5). This is important for it is related to the claims that paradox is an irrevocable trait of thinking, that philosophy requires friends both within and outside its rather blurry boundaries, and that reason cannot, in the end, claim self-grounding knowledge of its own origins and ends. Fain, like Rousseau, writes as a friend of the love of wisdom and “[a]gainst all the odds, the combination of Rousseau with Laplanche amounts to something of a double-dog dare against the ruling conventions of academic philosophy” (ix). The wager has been laid on the table, but it will extend far beyond the contemporary academy.A certain image of Socrates is the central figure around which Fain’s reading of Rousseau is organized. This is Socrates as that oddball Athenian irritant who works to demonstrate, succinctly put, that “virtue is knowledge; happiness is knowledge of the good” (4). This Socrates—and [End Page 92] Socrates is always multiplying—knows the irony of the unknowing and the joy of the virtuously beautiful. Phronēsis will be more foundational than epistēmē and philosophy, in itself—however that “in itself” is construed—will not be sufficient to explain itself. It has a constitutive blind-spot that will need looking into from different perspectives. It will need Rousseau and Laplanche, for instance, as external supports in order to get it into gear, with Laplanche being the psychoanalyst who most systematically brings this “support” of Freud’s Anlehnung—often translated as “anaclitic”—as a relation between the Instinkt of biology and the Trieb of the psyche, into sharper critical relief.Plato, in Theaetetus, succinctly states the origin of philosophy is “wonder” at a point in the conversation focused on questions of becoming and definition, which have already made the Theaetetus “dizzy.” (We all know what that is like.) Socrates replies to this confession, noting that “It looks as though Theodorus’s sketch of your character was accurate my friend. I mean, this feeling—a sense of wonder—is perfectly proper to the philosopher: philosophy has no other foundation, in fact. The genealogist in question is Hesiod, who, in the Theogony (265–6), observed that Iris (the rainbow as a ‘shining wonder’) was the daughter of Thaumas (‘wonder’) and Electra (‘shining’). Let me limit my comments here to simply observing that Theaetetus’s character, not just his ratiocination, matters. The dizziness becomes an opening to the shining wonder “beneath which” there is no other foundation.Socrates, as he so often does, translates the poetic into the philosophical, but without losing sight of the former. The mythopoetic—for both Plato and Rousseau—is inseparable from logic’s explanatory force and cannot be extirpated by that force. This relationship is active throughout Primal Philosophy and binds together the philosophical, the artistic, and the psychoanalytic. According to the state of the art in psychology and philosophy, colour sensations are located in a 'quality space'. This space has three dimensions: hue , saturation , and brightness. This space is structured further via a small number of primitive hues or landmark colours, usually four or six . It has also been suggested that there are eleven semantic universals — the six colours previously mentioned plus orange, pink, brown, purple, and grey. Against the standard view, we argue that (...) colour might better be regarded as the outcome of a social-historical developmental trajectory in which there is mutual shaping of philosophical presuppositions, scientific theories, experimental practices, technological tools, rhetorical frameworks, and their intercalated and recursive interactions with the lifeworld. That is: the domain of colour is the outcome of interactive processes of scientific, instrumental, industrial, and everyday lifeworlds. (shrink)

The Drosophila salivary gland has emerged as an outstanding model system for the process of organ formation. Many of the component steps, from initial regional specification through cell specialization and morphogenesis, are known and many of the genes required for these different processes have been identified. The salivary gland is a relatively simple organ; the entire gland comprises of only two major cell types, which derive from a single contiguous primordium. Salivary cells cease dividing once they are specified, and organ (...) growth is achieved simply by an increase in size of individual cells, thus eliminating concerns about the potential unequal distribution of determinants during mitosis. Drosophila salivary glands form by the same cellular mechanisms as organs in higher organisms, including regulated cell shape changes, cell intercalation and directed cell migration. Thus, learning how these events are coordinated for tissue morphogenesis in an organism for which the genetic and molecular tools are unsurpassed should provide excellent paradigms for dissecting related processes in the more intricate organs of more complicated species. The external surfaces of the human body, as well as its internal organs, constantly experience different kinds of mechanical stimulations. For example, tubular epithelial cells of the kidney are continuously exposed to a variety of mechanical forces, such as fluid flow shear stress within the lumen of th nephron. The majority of epithelial cells along the nephron, except intercalated cells, possess a primary cilium, an organelle projecting from the cell's apical surface into the luminal space. Despite its discovery over 100 (...) years ago, the primary cilium's function continued to elude researchers for many decades. However, recent studies indicate that renal cilia have a sensory function. Studies on polycystic kidney disease (PKD) have identified many of the molecular players, which should help solve the mystery of how the renal cilium senses fluid flow. In this review, we will summarize the recent breakthroughs in PKD research and discuss the role(s) of th polycystin signaling complex in mediating mechanosensory function by the primary cilium of renal epithelium as well as of the embryonic node. Clive’s slim Marguerite de Navarre: An Annotated Bibliography made pointedly clear the marginal position of (...) this author in the French Renaissance literary canon, and the relative obscurity of her Heptaméron (there were less than 200 entries dealing with her unfinished collection of stories that first appeared in 1558). The 500th anniversary of Marguerite’s birth in 1992 brought with it a remarkable outpouring of reflections on this neglected masterpiece. Critical Tales ranks among the best of such undertakings. Eclectic in its approach, the volume comprises fifteen articles by scholars whose analyses advance the reader’s appreciation of the Heptaméron in all “its inexhaustible complexity” (p. 279).As complex as Marguerite’s tales may be, Lyons and McKinley have done an admirable job making their volume’s contents as accessible as possible to the growing number of scholars interested in the work both as text and as important artifact in the history of the evolution of narrative. All the articles are in English (with three translated from French). Citations from the Heptaméron appear in French, but for each, the editors include the corresponding page numbers in the most accessible English translation. In addition to a selected bibliography, the volume provides a useful index.The volume’s introduction presents the dominant issues, as well as a sketch of each article’s vantage point. The editors then divide the book into three sections: “Generic Transformations and Graphic Transgressions” (with articles by Robert D. Cottrell, Hope Glidden, Marcel Tetel, Donald Stone, and Tom Conley); “Narrative Systems and Structures” (with articles by Michel Jeanneret, Cathleen M. Bauschatz, François Cornilliat and Ullrich Langer, McKinley herself, Philippe de Lajarte, and André Tournon); “Character and Community” (with articles by Daniel Russell, François Rigolot, Paula Sommers, and Edwin M. Duval). The epilogue that follows, by virtue of its length and quality, could easily be considered the sixteenth article in the collection. It shows how these heterogeneous pieces relate to issues of narrative and narrative theory.Just how radical an upheaval did the Reformation cause? The contributions bear witness to the major changes in ways of thinking and writing that played themselves out even in secular texts. The theological tensions caused by the evangelical movement are evident in all three sections of the volume, and the introduction will lead readers to the relevant essays.
