While outcomes for children with T-cell acute lymphoblastic leukemia have improved dramatically, survival rates for patients with relapsed/refractory disease remain dismal. Prior studies indicate that glucocorticoid resistance is more common than resistance to other chemotherapies at relapse. In addition, failure to clear peripheral blasts during a prednisone prophase correlates with an elevated risk of relapse in newly diagnosed patients. Here we show that intrinsic GC resistance is present at diagnosis in early thymic precursor T-ALLs as well as in a subset (...) of non-ETP T-ALLs. GC-resistant non-ETP T-ALLs are characterized by strong induction of JAK/STAT signaling in response to interleukin-7 stimulation. Removing IL7 or inhibiting JAK/STAT signaling sensitizes these T-ALLs, and a subset of ETP T-ALLs, to GCs. The combination of the GC dexamethasone and the JAK1/2 inhibitor ruxolitinib altered the balance between pro- and anti-apoptotic factors in samples with IL7-dependent GC resistance, but not in samples with IL7-independent GC resistance. Together, these data suggest that the addition of ruxolitinib or other inhibitors of IL7 receptor/JAK/STAT signaling may enhance the efficacy of GCs in a biologically defined subset of T-ALL.Leukemia advance online publication, 30 May 2017; doi:10.1038/leu.2017.136. The International Bill of Human Rights (IBHR) enjoys universal acclaim as the source of the best standards and definition of human rights. This paper argues that the IBHR is inspired by liberalism and harbours ambiguities that open the door to a neoliberal seizure of the rights agenda; and that this effectively destabilises the focus on the IBHR on socio-economic and community rights, and therefore its stated ideal of the equal value of all human rights. I argue that Kwame Gyekye's moderate (...) communitarian political philosophy affords a viable philosophical basis for justification of a more balanced conception of human rights than the theoretical basis of the IBHR does. Discussion of sexuality in African cultures has a long history, but since the 1990s ethical reflections on homosexuality on the continent have often degenerated into furors and provoked a spate of anti-gay legislation in several countries. Refutations of homophobic dispositions encounter as barrier a pervasive belief in African cultures, that childbearing for community replenishment is a cherished moral duty. Several philosophers consider these to be exaggerated inhibitions that unjustifiably impede social acceptance of homosexuality, and have proposed as a solution what (...) they consider to be self-justifying political-moral principles, that terminate in value-pluralistic ideas such as the acceptance of the equality of sexual orientations and vindication of the right to the freedom to choose and satisfy sexual desire. I question the adequacy of such self-justificatory normative principles and consider the solutions they proffer as depreciating the moral point of the African pro-natalist position. Consequently, I develop a moral argument grounded in the ontology of Kwame Gyekye’s moderate communitarian theory of personhood as the most persuasive justification for homosexuality. (shrink)

Debate concerning virtual reality is often drawn in terms of sharply defined dichotomies--for example, between "real" (or "actual") and "virtual," "authentic" and "inauthentic," and "natural" and "artificial." In this paper we offer an alternative approach by suggesting a conception of a virtual world that highlights a continuity and commonality with our sense of everyday reality. We accomplish this in part by an examination of the English picturesque garden as if it were a virtual world partially constructed out of ideas and (...) objects collected during travels to foreign lands on the Grand Tour. Such foreign travel transformed not only the English person's sense of self, but also altered the English landscape. We conclude that in one sense the "real" England is also a "virtual" reality. Donald Davidson holds that intentional concepts are not reducible to physical or dispositional ones. This is due, he claims, to the constitutive role of normativity in the principles that govern the application of intentional concepts. According to Davidson, the specific way in which norms of rationality and coherence are mobilised by our interpretative principles sets mental concepts off from those of the natural sciences. I agree with Davidson on the irreducibility of the mental. However, I show that irreducibility is due (...) not to the normative character of intentional concepts, but to holism and the flexibility of interpretative principles. I then consider three arguments that Davidson has put forth to support the irreducibility of intentional properties on the basis of their normative character, and show that none of them goes through. This paper aims to lay bare the underlying logical structure of early Buddhist moral thinking. It argues that moral vocabulary in the Pali Suttas varies depending on the kind of agent under discussion and that this variance reflects an understanding that the phenomenology of moral experience also differs on the same basis. An attempt is made to spell this out in terms of attachment. The overall picture of Buddhist ethics that emerges is that of an agent-based moral contextualism. This account (...) does not imply that the prescription for moral conduct differs according to class of agent, but rather that the correct description of moral experience does. Further it implies that the descriptions of the moral experiences of different classes of agent differ phenomenologically, rather than in terms of overt behavioral characteristics. While most of the discussion is centered on the distinction between ordinary persons and disciples in higher training, the paper concludes with a brief exploration of the problematic moral experience of the arahat. Analyses of preference for the timing of uncertainty resolution usually assumes all uncertainty to resolve in one point in time. More realistically, uncertainty should be modelled to resolve gradually over time. Kreps and Porteus (1978) have introduced an axiomatically based model of time preference which can explain preferences for gradual uncertainty resolution. This paper presents an experimental test of the Kreps-Porteus model. We derive implications of the model relating preferences for gradual and one-time resolving lotteries. Our data do not support (...) the Kreps-Porteus model but show that some of the behaviour observed may be explained by similarity heuristics. (shrink)

TWEEDALE On SEVERAL OCCASIONSin his logical writings Abailard extracts himself from embarrassing ontological implications of his analyses of language by resorting to the notion of a something that is not a thing. I shall note here two such occasions and then discuss Abailard's explanations of this procedure based on the grammatical distinction of personal and impersonal constructions. Since the texts on this latter (...) topic are especially confusing, a considerable portion of my effort will be spent on making sense of them as preparation for drawing some conclusions about Abailard's overall approach. My purpose here is to show how Abailard's grammatical analyses are fundamental to his defence of a number of metaphysical positions. I also want to suggest that Abailard's grammatical approach, especially on the question of dicta, contains insights of continuing interest. Not all of Abailard's non-things are particularly associated with analyses of language. For instance, there are the likenesses the mind supposedly constructs for itself in order to think of things that are not being perceived. These Abailard calls fictive things (res ficta), and he refuses to give them the status of real psychological entities. ~ The likenesses are compared to mirror images which Abailard is also willing to treat as non-things. However, I wish to restrict my discussion here to cases which arise from Abailard's analysis of the reference of certain words. One such instance occurs in his discussion of universals.
